Identify a species of wild macrofungi which was found in the Jinshan area of Fujian Agriculture and Forestry University, discussing a better method of microbiology identification. Method: Using the traditional identification methods from the morphology of spore, single colony, hyphal and fruit body; Using the methods of molecular biology to amplify and sequence the strains of ITS(Internal Transcribed Spacer)sequence, then BLAST it in EMBL, and construct the Phylogenetic tree of the ITS sequence with Clustal X1.83 Mega 4.0. Result: (1) Obviously there is septum in mycelium of the subject, and clamp connection can be seen easily. (2) The length of ITS sequence of the subject is 565bp. (3) It can be seen from Phylogenetic Tree that the subject and Tricholoma giganteum homology is very high, so these two species are both Tricholoma.(4)Preliminary identified the wild unknown strains belong to Homobasidiomycetidae Agaricales Tricholomataceae Tricholoma. We call it Tricholoma jinshanense temporary. (5) The morphological characteristics of large fungi with ITS sequence analysis can be used in the macrofungi classification and identification. Using orthogonal design L16 (44) were applied to research the best culture conditions of Tricholoma sp mycelia,including the best source of carbon、the best source of nitrogen,the optimum temperature and the optimum pH.The experiment results indicated that the optimum C–source for its mycelia growth was sucrose and the optimum N–source was yeast;The optimum temperature was 25℃;The optimum pH was 7.0.And the fruit body of Tricholoma sp on the contain of moisture, Polysaccharide, protein, fat and Amino are 83.66%, 33.15%, 28.41%, 4.23% and 13.97%.
