| Commonly the flower color of rape is yellow, heart leaf color is usually green, but also there are some Brassica napus L. resources with golden yellow, orange, white or pure white flower color or the heart leaf color is purple. Flower color and heart leaf color traits act as indicating traits,which have many advantages, such as stable performance, easy to distinguish, not affecting other agronomic traits etc, and can be used for the determination of natural hybrid rate, purity identification and detection of character transfer, etc. At present, the research on the orange flower color and heart leaf color traits of Brassica napus L.mostly stay in the production and genetic analysis, this study tries to find molecular markers tightly linked with orange flowers and purple heart leaf by AFLP combined with BSA method, which can be applied in molecular marker assisted breeding. In this study the genetic law of flower color and heart leaf color traits were studied by using orange flower, purple heart leaf rape and their separation populations for research materials, molecular markers tightly linked with orange flowers and purple heart leaf gene were found, and discusses how purple heart leaf influenced the production of Brassica napus L. The main research results are as follows:1. In the process of genetic research, the genetic rule of BC1 segregation population followed the separation ratioof 3:1,F2 segregation population followed the separation ratio of 15:1, the results Showed that orange flower trait was controlled by two pairs of recessive genes. By using molecular marker method combined with BSA, 10 AFLP markers which were closely linked with HS1 was found, and 7 specific segments were successfully sequenced. Finally contrast analyses of the results of the 5 segments indicated that the target gene was most likely located on the A9 chain group of Brassica napus L.2. In the process of genetic research, the genetic rule of BC1 segregation population followed the separation ratio of 1:1, F2 segregation population followed the separation ratio of 3:1, the results Showed that purple heart leaf trait was controlled by one pair of dominant gene. Refering to the methods of color traits, 10 molecular markers which were closely linked with ZX was found, including 8 AFLP markers and 3 SSR markers, ZXwas successfully located in 1.1Mb range of C6 chain group in Brassica napus L.3. In populations yield analysis of purple heart leaves and green heart leaves were made. The results showed that purple heart leaf trait had no significant effect on the yield of Brassica napus L. It was feasible to develop a purple heart leaf variety. The molecular markers closely linked with gene ZX can be used in molecular marker assisted breeding, also can be used in rape hybrid purity identification of Brassica napus L. |
