| Inner Mongolian fine local variety Ujumuqin sheep has its advantages and characteristics on adaptability, meat quality and disease resistance, etc. There is a lot of variation in genes which have a mix of adaptive significance and application value. Multi-vertebra phenomenon is Ujumuqin sheep’s most common production performance and unique characteristic.Ujumuqin sheep’s meat production is more than ordinary sheep; therefore they are generally praised and welcomed very much. Scientists and the majority of pastoralists pay more attention on them nowadays.In this study, we chose Hoxa11, Hoxd11 and Hoxc8 genes which affect multiple-vertebra as candidate genes, designed to explore the relationship between polymorphism of these three genes and phenotype of Ujumuqin sheep’s four different type (T13L6, T13L7, T14L6, and T14L7), provided further scientific evidence for developing genetic study of Hoxa11, Hoxdlland Hoxc8, and laid the foundation for cultivate our meat sheep breeds.In this experiment, we used 40Ujumuqin sheep as the research object, which contains 10 Ujumuqin sheep without multiple-vertebra (T13L6),10 multiple-lumbar Ujumuqin sheep (T13L7),10 multiple-thoracic Ujumuqin sheep (T14L6),10 multiple-thoracic and lumbar one. According to published Texel sheep Hoxall, Hoxdll and hoxc8 gene’s cDNA sequences designed the primers and amplified all three gene exon sequences. The use of DNA sequencing and sequence analysis, and the method of molecular biology, we studied Hoxa11, Hoxd11 and hoxc8 genetic polymorphism.Results showed as follow:(1) Ujumuqin sheep Hoxa11, Hoxd11 and hoxc8 genes entire exon were 6519bp,1320 bp and 2188 bp. Hoxa11 gene contains 5 exons,2449bp of CDS region,4442bp of 5’UTR region, and 1252bp of 3’UTR region.(2) Ujumuqin sheep Hoxd11 gene contains 3 exons, CDS full-length was 473bp,847bp of 3’UTR region.(3) Ujumuqin sheep Hoxc8 gene contains 3 exons,62bpof 5’UTR region,1532bp of 3’UTR region and 729bp of CDS region.(4) This study enriched the Ujumuqin sheep Hoxa11, Hoxd11 and Hoxc8 gene sequences,and the sheep of the three genes research provides the experimental basis. |
