Soybean is one of the important crops in China for protein and oil production. It plays an indispensable role in agriculture. Soybean varieties torelant to herbicide developped by genetic engineering has been widely used worldwide. Cultivation of herbicide-resistant soybean can not only reduce the labour cost for weed management and yield loss.In the study, three herbicide-resistant transgenic soybean lines, ZUTS-31, ZUTS-32 and ZUTS-33, which were achieved by agrobacterium mediated transformation. The transgene used for the study is a g10-epsps gene isolated from Deinococcus radiodurans. The recipient soybean cultivar is Huachun No.3. The herbicide-resistant transgenic soybean lines were produced through the expression of exogenous g10-epsps protein which has no affinity site of glyphosate herbicide. It could substantially improve efficient of field weed control reduce seed production cost.Southern blot analysis showed that the exogenous g10-epsps gene was successfully inserted in the soybean genome with a single copy in all the three transgenetic lines. Western blot analysis showed that the objective protein was highly expressed in the different developmental stages of plant roots, leaves and other tissues.Three insertion sites of T-DNA in the three transgenetic lines were obtained by TAIL-PCR. In the lines of ZUTS-31,32 and 33, T-DNA was inserted in the soybean 15th,13th and 19th chromosome, respectively. The inserted T-DNA included the region from the right boarder to the left boundary in the binary vector pSOY19.Using RT-PCR, Western blot and ELISA analysis, the abundance of the g10-epsps proteinin the different generations of the transgenic lines were analyzed. Result showed that the transgene was expressed in all three transgenic lines.In summary, the study accomplished the molecular characterization of the three transgenic soybean. The data is essential for the safe assessment of the transgenic lines.
