The piglet diarrhea is a disastrous intestinal disease that effect survival, development and growth. To varying degrees, the pigelet diarrhea was prevalent in different swine farms. As different places and diversifications of mangerment and sanitary condition, the pathogenic bacteria and virus are different and the pathogenicity is also different. So the pathogenic bacteria and virus as well as their pathological and biochemical characteristicsin different swine farms of Gansu and Henan were studied.The amplification technology of 16 S rDNA was used to identify bacterial etiology. The serotype of enterotoxigenic E.colic was confirmed by standard diagnostic serum and drug resistance analysis of bacterial etiology was performed using disk diffusion test. The results showed that the pathogenic E.colic was isolated in 80 samples, Salmanella and E.colic were isolated at the same timein 8 samples; The advantage serotype of enterotoxigenic E.colic was O78:K80(B) and the pathogenic E.colic had appeared high resistance.The 262 feces samples of diarrheawere detected by PEDV(procine epidemic diarrhea virus)-TGEV(transmissible gastroenteritis of swine virus)-PRV(porcine rotavirus)triplexreal-time fluorescent RT-PCR kit.The positiverates of PEDV, TGEV and PRVwere 48.1%, 1.5%, 19.1%respectively. The RNA of PEDV was extracted from positive fecal samples and amplifiedSgene. Then,the phylogenetic relationships of PEDV strains were constructed complete based complete Sgene.The results suggested that the genetic distance is close in PEDV strainsisolated from different swine farms in the same region, and the S gene of PEDVdisplay multiformityin different regions.The levels globulin, urea and creatinine in serum of diarrheal piglets were increased significantly(P<0.05)by comparing the blood biochemical indexes of PEDV-infected piglets and healthy piglets.The result suggested that the functions of protein metabolism, liver and kidneys may be impaired by PEDV usingPrincipal Component Cluster analysis method.With comparing the pathological section of PEDV-infected piglets and healthy piglets, we found there were a large amount of inflammatory cells and the intestinal villus was atrophic in small intestine of PEDV-infected piglets.In the liver and kidneytissue of PEDV-infected piglets, the hepatocyteswerepartial denatured and necrotic, and renal tubular epithelial cellswere deciduous andnecrotic.In addition, the PEDVcaused IKKβexpression level increased in the ileum of piglet by comparing IKKβexpression level of different intestine segments.The study concluded that the serotypeO78:K80(B) and PEDV were the major pathogeny caused of pigletsdiarrhea in Gansu and Henan province, and the genetic distance of PEDVwas close in the two regions. Besides could damage tothe intestine function of piglet and increaseIKKβexpression level, the PEDV could cause protein metabolism disorders and dysfunction of kidney and liver.
