| Heat shock proteins(HSPs) are rapidly synthesized in response to environmental stressors, which are closely related to the tolerance at high and low temperatures. HSPs play important roles in the growth and development of insects. Here, we devoted to investigating the thermal and cold tolerance of Dendroctonus armandi and responding for high and low temperatures, Furthermore, we explored the expression profiles of heat shock proteins for short-term treatments, in order to indicate the molecular mechanism on thermal adaptability of Chinese white pine beetle, and provide reasonable measures for prevention and control at meanwhile. Main results of this study are shown as follows:1. We have cloned 16 heat shock protein genes from Chinese white pine beetle(8 of Hsp70 famLiy, 2 of Hsp90 famLiy, 6 of sHsp famLiy), which were uploaded to genebank and got accession numbers.2. The nucleotide sequences of the Hsps are highly similar with some other insect species,and highly conserved in most organisms.3. Hsp70, Hsp90 and sHsp showed up-regulation changes in responding to cold and heat hardening, and their expression levels increased with the temperature rising and reducing when reaching the maximum. The expression levels of Hsp70 were significant in high-temperature stress, and those of sHsp were significant in low-temperature stress.4. The sensitivity for temperature of the females is greater than the male, while, male can adapt wider temperature range than female. Furthermore,we found that the expression level of DAHsp70-8 was up-regulated only in female. This result indicated that DAHsp70-8 is closely related to female beetles. |
