| Marek’s Disease (Marek’s diseases, MD) is one of serious epidemic Diseases that does harm to healthy development of the poultry industry, which can cause high morbidity and mortality in chickens and lead to significant economic losses in the poultry industry. At present, although the application of vaccine on the prevention of the disease has certain effect, but the immune failure and coupled with the strong virus strains and Super virus strains Marek’s disease virus (MDV) appears bring some new difficulties in preventing and treating of the disease. Therefore, the effective preventive measures is one of the important parts to prevent the occurrence of MD. Chinese herbal medicines have a long history, and it is rich in resources, with no residue, no resistance, and can improve the advantages of animal production performance and disease resistance and so on. Thus in the livestock and poultry breeding industry as feed additives has become one of the hot researches. The study intends to screening of four kinds of Chinese herbal medicine compounds and apply chicken MD animal model to research the effect of Chinese herbal medicine compounds on anti-virus, according to antioxidant indexes and cytokines to research the effect of Chinese herbal medicine compounds on the immune function and anti MDV in chickens, and for the Chinese herbal medicine compounds as feed additive in anti-MD virus and enhance immunity of the application to provide a scientific basis for reference.At first, screening of Chinese herbal medicine compounds with immune enhancement. Choosing one day old healthy chickens and randomly divided them into 5 groups, the first group is control group, only feed basal diet. The other four groups are the experimental groups, Chinese herbal medicine compounds are added to the basic diets. Four kinds of Chinese herbal medicine are mostly contain Dangshen, Lianqiao etc and supplemented with two different functional amino acids. According to the pathogenesis stages of MD, at 21,35 and 42 day old, measuring antioxidant indexes of serum and the levels of cytokines in chicken. The analysis of the indexes shows that:compared with the control group,the groups which added four kinds of Chinese herbal medicine compounds’serum T-SOD activity have trend of enhancement,at 42 day old the experimental group which added Chinese herbal medicine compounds four, its T-SOD activity significantly enhance. The MDA concentration have trend of reduction, the experimental group which added Chinese herbal medicine compounds four, its MDA concentration significantly and extremely significantly decrease(P<0.05,P<0.01). At 35,42 day old the experimental groups which added Chinese herbal medicine compounds three and four, its concentration of serum interleukin 2 (IL-2)ã€interleukin 18 (IL-18)ã€Tumor necrosis factor alpha (TNF-a) and interferon gamma (IFN-y) are significantly higher than the control group (P<0.05), but its concentration of serum interleukin 6 (IL-6), interleukin 8 (IL-8), interleukin 10 (IL-10) are significantly or extremely significantly lower than the control group at three different period. The four kinds of Chinese herbal medicine compounds to add feeding, but effect of different day old chicken was different. The T-SOD activity and MDA concentration of groups which added four kinds of Chinese herbal medicine compounds are not significantly different (P>0.05). The test groups which added Chinese herbal medicine compounds three and four,its concentration of serum IL-2, TNF-a and IFN-y in the 42 days of age are significantly higher than 21 day old, and the concentration of IL-6, IL-8, IL-10 compared to 21 day old are significantly and extremely significantly decrease (P< 0.05, P< 0.01).Through the comprehensive analysis of two different aspects of discovery, it show that the four kinds of Chinese herbal medicine compounds adding based diets can improve the immune functions of chickens, but the effect of Chinese herbal medicine compound four is the most significant.Secondly, application four kinds of Chinese herbal medicine compounds as additive to study its effect of anti-MDV. Choosing one day old healthy chickens and randomLy divided into 6 groups,1 groups are set as control group, each one inject normal saline 0.5mL into abdominal cavity. The other five groups are the MDV-infected group, each one inject 0.5mL/MD blood poison into abdominal cavity to establish MD animal mode.The MDV-infected control group and the control group are feed the based diet, the MDV-infected experimental group feed based diet that adding four kinds of Chinese herbal medicine compounds. At 21,35,42 day old, measuring antioxidant indexes of serum and the levels of cytokines in chicken respectively. Statistical results show that, Compared with the control group, the MDV-infected group that addition Chinese herbal medicine compounds reduces the morbidity and mortality of chickens. Analysis results of MD animal model chicken serum antioxidant indexes and cytokines results show that, the MDV-infected control group compare to the control group, its T-SOD activity is significantly decrease, the concentration of MDA is significantly increasing (P<0.05), at different day old, IL-2, IL-18,IFN-y and TNF-a concentration are significantly or extremely significantly lower than control group (P<0.05, P<0.01), the concentration of IL-6, IL-8 was significantly higher than control group (P<0.05). Compared with the MDV-infected control group, the groups of adding compound Chinese herbal medicines’ T-SOD activity have trend of enhancement, at 35 day old the experimental group which added Chinese herbal medicine compounds four,its T-SOD activity significantly enhance(P<0.05).. The MDA concentration have trend of reduction, the experimental group which added Chinese herbal medicine compounds four, its MDA concentration significantly decrease (P<0.05). At 35,42 day old, the MDV-infected experimental groups which added Chinese herbal medicine compounds three and four., it’s concentration of serum IL-2, IL-18, IFN-γ and TNF-a significantly or extremely significantly higher than the MDV-infected control (P<0.05,P<0.01), At 42 day old, the MDV-infected experimental group which added Chinese herbal medicine compounds three and four, its concentration of serum IL-6, IL-8,IL-10 significantly lower than the MDV-infected control (P<0.05). The results show that the Chinese herbal medicine compounds can improve the anti-viral infection ability of chickens, and the analysis results show that the effect of the Chinese herbal medicine compound four is the best.The research results show that add four kinds of Chinese herbal medicines in chicken diet can improve the body’s antioxidant ability in different degrees, raise the body’s immune function and anti-viral ability.The function of Chinese herbal medicine compound four is the best. The study result provides valuable reference for Chinese herbal medicine compounds as feed additives and further application in the poultry disease prevention. |
