Porphyra yezornsis(Bangiales, Rhodophyta)is one of the most extensively cultivated species in China, and there are many researches that related to the breeding, physiological and biochemical of it.The sexual differentiation is an important process in biology of development. Such study is benificial from the suitable experimental materials. Since the macroscopic foliage gametophyte of P.yezoensis is composed of single cell layer and the spermatangium is developed from the vegetative cell and distributed in the edge of the blades. This process is analogous to the development of the microspore to the spermatia in the higher plants. Porphyra sp. are amongst the best candidates in algae and higher plants used to study the sexual differentiation process.In this article, collected the vegetative cells and antheridium cells from P.yezoensis with cold water to maintain its good physical activity. Then observe the materials with the microscope, it shows that the vegetative cells was irregular quadrilateral, and each Antheridium cells divided into 2-8 parts. Both materials collected for 2g and then quickly frozen in liquid nitrogen, the phenol extraction method was choosed as the best way to extract the total protein from P.yezoensis in the event that there are many interfering substances of salinity, polysaccharide and pigments in P.yezoensis and it is positive for the subsequent mass spectrometry experiments. After detecting the extracted protein with Two-dimensional electrophoresis, analyze the differences between them by ITRAQ. It shows that 549 different proteins are found in both materials,101 stable protein were obtained after screening. Comparing with vegetative cells,88 protein up-reg ulate and 13 protein down-regulate in Antheridium cells. Among these proteins 31 protein involved in proteins translation,29 protein related to photosynthesis,7 protein associated with ATP synthesis,9 protein participated in fatty acid synthesis et al. We choose proteins that related to protein synthesis, carbon cycle, ATP synthesis for real-time PCR and found that actin, HSP 90, HSP 70-2, cytochrome f, calmodulin down-regulate and tubulin and histone H2B up-regulate in antheridium cells when comparing with vegetative cells. The result showed that HSP 70-2, calmodulin were oppsited to ITRAQ, the others were identical.
