| 1-day-old SPF chickens were used in this study, applying the methods of TUNEL, HE staining, real-time fluorescence quantitative PCR, HIC, indirect ELISA and other methods, according to detected the numbeis of apoptotic cells, nuclear cytoplasmic ratio, Bcl-2 and C-myc gene expression levels and other related indicators, we studied the influences of the values above mentioned in immune organs(thymus, spleens and bursa of Fabricius) of SPF chickens infected with REV comprehensively and systematically. The study aims to provide important experimental evidence for the mechanism of RE occur and its prevention. These results revealed that:(1) The number of apoptotic cells in the immune organs of 1-day-old SPF chickens infected with REV is higher than that in control chickens in varying degrees, and especially in bursa of Fabricius. Among them, 21~28 days later after infected with REV, the number of apoptosis cells in thymus and bursa of Fabricius is significantly(P<0.05 or P<0.01) higher than that in control chickens. At 42 d, the number of apoptotic cells in bursa of Fabricius remained significantly(P<0.05) higher than that in control chickens. The nuclear cytoplasmic ratio in immune organs is less than that in control chickens in different degrees a fter chickens were infected with REV. And the nuclear cytoplasmic ratio in bursa of Fabricius is significantly(P<0.05) less than the control chickens after infected 21~28 d. Although the nuclear cytoplasmic ratio in thymus and spleen are less than that in control chickens, there is no significantly(P>0.05) changes.(2) The Bcl-2 positive cells number, Bcl-2 mRNA and protein expression levels in the immune organs of 1-day-old SPF chickens infected with REV are higher than that in control chickens in varying degrees. Among them, 21 days later after infected with REV, the number of Bcl-2 positive cells in thymus and bursa of Fabricius is significantly(P<0.05) higher than that in control chickens. At 28 d, the number of Bcl-2 positive cells in thymus and spleen increase obviously(P<0.05) to compare with that in control chickens, but the number of Bcl-2 positive cells in bursa of Fabricius is very significantly(P<0.01) higher than that in control chickens; the Bcl-2 mRNA expression and its protein content in thymus, spleen and bursa of Fabricius are obviously(P<0.05 or P<0.01) higher than that in control chickens after infected 21~28 d.(3) The C-myc positive cells number, C-myc mRNA and protein expression levels in the immune organs of 1-day old SPF chickens infected with REV are different degrees higher than that in control chickens. Among them, the number of Bcl-2 positive cells in thymus and bursa of Fabricius is significantly(P<0.05) higher than that in control chickens after infected respectively 21 d and 21~28 d. C-myc protein content in thymus is significantly(P<0.05) higher than that in control chickens after infected 21~28 d. C-myc protein content in spleen is significantly(P<0.05) higher than that in control chickens after infected 28 d. C-myc protein content in bursa of Fabricius is not only very significantly(P<0.01) higher than that in control chickens, but also significantly(P<0.05) higher than the control chickens after infected 42 d. The Bcl-2 m RNA expression in the above mentioned three kinds of immune organs was significantly(P<0.05) or very significantly(P<0.01) higher than that in control chickens after infected respectively 21~28 d and 28 d. |
