Xanthoceras sorbifolia,Bunge.is good unique woody oil species in China,its seed oil content is 30% to 47%, kernel oil content as high as 66% to 70%, has the high industrial value and nutritional value, and contains a lot of unsaturated fatty acid, edible can prevention and treatment of atherosclerosis, is the important Chinese traditional medicine in the treatment of cardiovascular diseases, also provides the development and application of biodiesel raw material.Wen crown fruit has higher art characteristics at the same time, can adapt to low temperature, drought, poor soil, root developed, can absorb the deep soil moisture and nutrients, is the first selection of hill afforestation tree species.So the adversity stress below crown fruit of defense mechanism is of great importance.This study through to the condition of low temperature below crown fruit seedlings transcriptome analysis, find out the response of metabolic pathways and key genes in low temperature stress, the analysis of related hormone signal transduction pathways and its role in regulating under low temperature stress, the relative energy metabolic reactions variations,preliminary to clarify the molecular regulation mechanism of crown fruit seedling resistance to cold, specific results are as follows:1.Adoption of the transcriptome differentially expressed genes in GO(Gene Ontology)enrichment analysis, including molecular function(Molecular Function), biological processes(biological process) and cells(cellular component) in three parts. Cellular components include: Golgi, thylakoid membrane, cell wall, nucleus, etc., of biological processes including synthesis of secondary metabolites, histidine methylation, chromatin condensation, abscisic acid signal activation process, oxidation of fatty acid metabolism superoxide radical scavenging, allyl signal transduction processes, etc., molecular functions including calmodulin-dependent protein kinase activity, sucrose transmembrane transporter activity,phenylalanine transaminase activity, hydrolase and oxidoreductase activity and so on.2. Enrichment analysis of transcriptome genes differentially KEGG(Kyoto Encyclopedia Of Genes And Genomes), found that fructose, mannose metabolic pathways, flavonoid biosynthesis, photosynthesis metabolic pathway, the glycolytic pathway, phenylalanine metabolic pathways, sucrose, starch biosynthesis, hormone signal transduction pathways,unsaturated fatty acid synthesis pathway, and so on.3. Use the MISA software filter sorbifolia transcriptome sequencing acquired 61,659 pieces Unigene, were detected in 8,749 SSR loci, identified six types of SSR: Single nucleotide(Mono-nucleotide) repeat SSR, double-base( Di-nucleotide) repeat SSR,three-base(Tri-nucleotide) repeat SSR, four base(Tetra-nucleotide) repeat SSR, five bases(Penta-nucleotide) and six-base repeat SSR(Hexa-nucleotide) repeat SSR. SSR markers to identify Primer3 using primer design.Through the above analysis of differentially expressed genes, we found that at low stress,sorbifolia flavonoids seedling gene expression was significantly associated with upregulated genes associated membrane system also changed significantly, while energy metabolism associated with increased expression of oxidoreductase Meanwhile, we found that low temperature stress, salt stress induced defense pathways are also up-regulated, indicating that sorbifolia, the salt stress and low temperature stress have a common defense mechanisms and metabolic pathways.These results provide an important basis for further reveal sorbifolia seedling chilling mechanism, analysis of key genes involved in cold-induced injury, and sorbifolia breeding cultivated under stress conditions. Unigene information sorbifolia transcriptome sequencing produced can be used as an effective source of development of SSR markers, obtained large quantities of SSR markers for genetic sorbifolia its sibling species diversity analysis,marker-assisted breeding, genetic mapping provided more abundant and reliable marker selection.
