| In order to supervise the illegal use of Clenbuterol-HCL(CL) in Chengdu’s swine industry, it’s necessary to select the best kind of card in quality, which is accurate and effective in detecting field, and to establish the detection scheme for frontline work of CL supervision.The methods of detection on CL residues from swine urine and tissue by Liquid-Chromatography-Tandem Mass-Spectrometry(LC-TMS) and ELISA was established in this study for means of result-identification; The detection results of the four cards used in Chengdu were compared. The main quality factors of detection card, including the threshold value, the rate of fake CL-positive, fake CL-negative,the accuracy, and influency factors were tested. The best kind of detection card, matched with detecting scheme was selected for the CL fast detection in field and the government’s supervision. The study results were showed below:The methods of detection on CL residues by Liquid Chromatography-Tandem Mass-Spectrometry(LC-TMS) was established. The separation was performed on a BDS-C18 column with gradient elution using methanol and 0.1% formic acid at a flow rate of 0.25 mL/min. Under multiple reaction monitoring mode(MRM), the effects of MS parameters, such as capillary voltage and collision energy were investigated and optimized. Under the optimized conditions, the detection limits of CL reached 0.1μg/L, the correlation coefficient was 0.9991. The standard curve of ELISA was made as Y=-24.769x+53.724, LOD was 0.3μg/L, the correlation coefficient was 0.9984.There was no significient difference of the CL residues detection result between ELISA method and LC-TMS method,except on the concentration of CL below 0.5 ng/mL.The threshold value agreed with detecting limit of all 4 kinds of cards, and the rate of fake CL-negative was 0%. The fake CL-positive rate of Weideweikan was significiently lower than that of other three(P>0.05). All samples, contained CL residues with the concentration below the detecting limit, could be detected by Weideweikan. The other tests on quality factors showed that the Weideweikan card was accurate, sensitive and effective on CL-detecting. The best temperature for detecting work was above 20℃. The Weideweikan card was the best kind of testing card for the CL rapid screening detection and supervision in field, combined with the LC-TMS identifying, if suspicious CL-positive sample was detected. |
