Extraction of active ingredients of 12 taste traditional Chinese medicine were in vitro antimicrobial activity to Salmonella pullorum. By 12 herbs using ultrasound, ethanol reflux, water bath heating method of extracting the active ingredients, the active ingredient in traditional Chinese medicine extracted from in vitro inhibition of Salmonella pullorum test, measure the active ingredients of traditional Chinese medicine for Salmonella pullorum minimum inhibitory concentration (Minimal inhibitory concentration, MIC) and minimum bactericidal concentration (Minimal bactericidal concentration, MBC), and the active ingredients of traditional Chinese medicine combined inhibition was determined by checkerboard method for Salmonella pullorum effect. The results showed that:Burnet, forsythia, Polygonum strong inhibitory effect MIC are 1.95-15.6mg/ml, MBC was 3.9-15.6 mg/ml; Andrographis, Prunella, Terminalia MIC was 31.25 mg/ml, MBC is 31.25 mg/ml; Polygonum, Viola, bulbul, gutta, Bupleurum MIC is 62.5 mg/ml, MBC was 62.5-125 mg/ml; combined antibacterial test results showed that:Polygonum cuspidatum, myrobalan extract on Salmonella pullorum has strong antibacterial activity in vitro. Terminalia and Polygonum cuspidatum, Viola, Polygonum. Prunella showed synergistic antimicrobial joint; knotweed and Viola, Prunella, Viola and Prunella combined inhibitory effect was irrelevant; Polygonum cuspidatum and, Viola Prunella was combined inhibitory antagonism. Compatibility relations flavors of traditional Chinese medicine, for the development of anti- Salmonella pullorum compound Chinese medicine foundation. The results showed that: medicines knotweed eluent petroleum ether (30～60℃)-ethyl formate-formic acid (15:5:1), Chinese herbal medicine Terminalia developing solvent toluene-ethyl formate-formic acid (2:7:1) as the TLC plate sprayed ferricyanide reagent, medicines Viola developing solvent toluene-ethyl acetate-formic acid (4:3:1); to extract rate. MIC and MBC for the evaluation of its optimum extraction are:Polygonum cuspidatum optimum extraction of 60% ethanol. solid-liquid ratio of 1:5. extraction temperature is 95 ℃, each 1.5h, Terminalia optimum extraction process is expected liquid ratio of 1:20. extraction temperature was 100℃ . extracting two times, each time 1.5h. optimum extraction Viola extracted as follows:60% ethanol, solid-liquid ratio of 1:20, soaking 24h, ultrasonic time 60min. Conclusion:Polygonum cuspidatum, Terminalia and Viola meets Pharmacopoeia, determine the optimum extraction process shamisen medicine.3. "Huhezi" granule is preparation and quality control. Orthogonal design method accessories " Huhezi " granules:a-lactose, soluble starch and dextrin were screened in different proportions, HPLC and UV spectrophotometer with granules and Terminalia total polydatin polyphenol content was determined; results showed that:the best ratio of traditional Chinese medicine and accessories 1:4 liquid chromatographic conditions acetonitrile-water (23:77) as the mobile phase: column temperature was 30℃:flow rate was 1.0mL·min-1, polydatin concentration in the 20μg· mL-1～70μg·mL-1 and the peak area was a good linear relationship, gallic acid as reference in 53.94μg·mL-1～125.86μg·mL-within a range with a good linear relationship between absorbance. Conclusion:According to the orthogonal test materials granules of uniform size, water content, clarity meet the requirements and the range of the predetermined quality standards.4. To investigate drug compound " Huhezi," the toxicity and efficacy of Salmonella pullorum. Dose greater than 5000mg/kg, no death without any response in mice that LD50> 5000mg/kg. its traditional Chinese medicine-Determination of the maximum tolerated dose of the extract the maximum tolerated dose of 40g/kg; higher doses particles agent group effective rate and mortality rates were 83.3% and 23%,middle does group effective rate and moilality rates were 81.8% and 25%, low-dose group effective rate and mortality rates were 44% and 36%.the control group mortality was 40%,in the three granules of relative growth of 92.94%,90.9%,89.3%, western group relative growth of 85.98%.Conclusion:Chinese herbal compound is non-toxic; high dose group died on Salmonella putherum were effectively controlled drags for clinical application of compound enough to mention a theoretical basis. |
