| According on clinical symptom, pathological change and etiopathogenesis of chicken Newcastle desease, Five Chinese herbal medicinal prescriptions were formed according to the theory of based treatment based on syndrome differentiation. Therapeutic effects of the 5 Chinese herbal medicinal prescriptions were observed by artificial infection NDV to screen better Chinese herbal medicinal prescriptions, and studied their effects of anti-Newcastle desease and the changes of immunity endocrine indexes. When the main immunity endocrine changes of anti-Newcastle desease effect of these new prescriptions were approached, a Chinese herbal medicinal authenticate prescription according to the main mechanisms was formed. Finally, the anti-Newcastle desease effects of the authenticate prescription and the changes of immune-endocrine net and blood physio-biochemical indicators in these chickens treated by the authenticate prescription were observed in the present experiment. The present study contained the following 4 parts:Test I Preliminary screening of Chinese herbal medicinal prescription (CHMP) anti-ND In order to screen CHMP of anti-ND and to enhance its clinical therapeutic effect anti-ND, five new CHMPs were formed based on the etiopathogenesis of ND. Two hundred and thirty-nine chickens were assigned to seven groups:five CHMPs groups, NDV control group and blank control group. When infected artificially by NDV, the incidence of chickens was observed with the purpose of screening prescriptions. The results suggested that five CHMPs could delay initial time of invasion, weaken the injury of organism and decrease mortality when compared with NDV control group. CHMP-1 and CHMP-2 showed the best effect in significantly delaying invasion hour, lightened the bleed of glandular stomach and duodenum, and raising the survival rate of chicken infected by NDV. CHMP-2 was slightly better than CHMP-1. The effects of CHMP-1 and CHMP-2 were better than those of CHMP-4, CHMP-3 and CHMP-5. It was indicated that CHMP-1 and CHMP-2 could be used as preliminary screening prescription anti-ND.Test Ⅱ Effect of Preliminary screening prescription on endocrine immunity regulation in chickens infected with NDV artificially To approach the mechanism of endocrine-immunity of Preliminary screening prescription anti-ND, one hundred ninty-six chickens were divided randomly into five groups:CHMP-1 (Qi Lanyin) group, CHMP-2 group and Vaccination control group, NDV control group, Blank control group. When artificially infected with NDV, the proliferation of chicken peripheral blood lymphocyte (A570), serum antibody titers, and concentrations of IL-2, IFN-γ, IL-6, T4 and Glucagon in serum were determinated. The results showed that on initial time of invasion, compared to NDV control group, it was significantly raised for proliferation of chicken peripheral blood lymphocyte(A570), serum antibody titers and concentrations of T4 in Vaccination control group, serum values of T4 and IFN-γ in CHMP-1 group, and serum antibody titers and serum levels of T4, IL-6 and IFN-γ in CHMP-2 group. Serum antibody titers and serum concentrations of T4 in CHMP-2 were significantly higher than those of CHMP-1 group. On peak stage of leathal, compared to NDV control group, it was significantly raised for proliferation of chicken peripheral blood lymphocyte(A570), serum antibody titers and serum values of IL-2, T4 and glucagon in vaccination control group, however decreased significantly for serum IL-6 values in groups vaccination control, CHMP-1 and CHMP-2. When compared to CHMP-1 group, serum concentrations of T4 in CHMP-2 group were raised significantly, serum levels of IFN-y were increased slightly. The results indicated that specific immunity regulation was the main mechanism for immunized chicken anti-ND. CHMP-1 and CHMP-2 could promote specific immune response of un-immunized chicken infected by NDV, and the effect of CHMP-2 was better than CHMP-1. The anti-ND effect of CHMP-1 was partially owing to the quick promoting of serum IL-2 and IFN-γ in un-immunized chicken; however, CHMP-2 was partially owing to the quick upgrading of serum IFN-y and IL-6. Moreover, the anti-ND effect of CHMP-1 and CHMP-2 were possibly related to keeping un-immunized chicken energy metabolic balance via serum T4 regulationTest Ⅲ Effect of Chinese herbal medicinal authenticate prescription anti-ND and study on the change of immune-endocrine index in chicken In order to observe the effect of Chinese authenticate prescription anti-ND and the changes of immunity endocrine regulation in chicken, one hundred and fifty-two 30-day-old immunized chickens were randomly divided into five groups:Group I(prescription 1), Group Ⅱ(prescription 2), Group Ⅲ (Prescription 6-Chinese authenticate prescription), NDV control group and Blank control group. When artificially infected with NDV, the incidence of chicken was observed. At the time of before infected, leathal peak hour and stationary phase, six blood samples from each group were collected randomly from brachial vein for determination of proliferation of chicken peripheral blood lymphocyte (A570), serum antibody titers, and concentrations of IL-2, IFN-γ, IL-6, T4 and glucagon in serum. The results showed that Chinese herbal medicinal prescription could delay invasion time, relieve patho-injury, promote rehabilitation and decrease mortality of chicken effectively. Chinese herbal medicinal authenticate prescription showed the best effect. Prescription 1 and 6 could quickly increase serum antibody titers. Prescription 1 could promote the concentrations of IFN-y and T4 of serum. Prescription 2 might raise the levels of IL-2 and IFN-y of serum. Prescription 6 could raise significantly the concentrations of IL-2, IFN-y and T4. The three Chinese herbal medicinal prescriptions could reduce the concentrations of IL-6 and glucagon during lethal peak hour. The effect of prescription 1 adjusting the secretion of IFM-γ and IL-6 was better than that of Prescription 6, but was weaker in adjusting the secretion of IL-2 and glucagon. The results indicated that Chinese herbal medicinal could stimulate quickly humeral-mediated immunity response in vivo of immunized cocks and resist NDV infecting by adjusting immune-endocrine channel, such as the levels of cell factors:IL-2, IFN-γ and IL-6, as well as the levels of endocrine hormone:T4 and glucagon and so on. In general evaluation the efficacy of Prescription 6 was better than that of the other two prescriptions.Test IV Effects of Chinese herbal medicinal authenticate prescription on blood physiological and biochemical of chickens infected with Newcastle disease virus To investigate the effects of Chinese herbal medicinal authenticate prescription (CHMAP) on blood physiological and biochemical of chickens infected with Newcastle disease virus,120 34-day-old healthy chickens were randomly divided into 3 groups:CHMAP group, virus group and negative control group. Chickens in CHMAP group were drunk with CHMAP at the dosage of lml per chicken for 3 days after 24 h artificially injected Newcastle disease virus.6 chickens in each group were randomly chosen and blood samples were collected by cardio puncture on days 0(D0),6 (D6) and 13(D13) after Newcastle disease virus injected. Blood physiological items including hemoglobin(Hb), erythrocyte sedimentation rate(ESR), red blood cell(RBC), white blood cell(WBC) and differential count(DC), and blood biochemical indices including total protein(TP), albumin(ALB), globulin(GLO), creatine kinase(CK), lactate dehydrogenase(LDH), alanine aminotransferase(AST), amylase(AMY) and urate(UA) were measured. The results indicated that the values of LC, Hb, TP, GLO and LDH at days 6 in virus group were significantly decreased, and the values of eosinophile granulocyte (EGC), and mononuclear were significantly increased as compared with that of negative control group at the same sampling time. On days 13, values of blood physiological and biochemical items aforesaid in virus group were near to that of negative control group (except EGC). While, the differences of the aforesaid blood physiological and biochemical items in CHMAP group were significantly lower than that of between virus group and negative control group. In conclusion, CHMAP could lessen the damage in physiology and biochemistry induced by Newcastle disease virus infection. |
