GLP Laboratory Animal Research Facilities Barrier Environmental Disinfection Methods

This study chooses the 84-disinfectant, benzalkonium bromide, Baidusha disinfectant as the research subjects which are the disinfectant representatives containing chlorine, quaternary ammonium salt and are frequently used in regular disinfecting work in the Shandong Provincial Institute Drug Safety Evaluation Center. Immersing, wiping and spraying methods, as the most common ways to disinfect, were tested to identify the effects in the GLP laboratory barrier environment with different densities and functioning durations. The target is to find the proper disinfectants and the effective methods of usage respectively. The results were obtained by experiment as following:These three kinds of disinfectants are all suitable for the immersion disinfection of the experimental animals feeding apparatus(rat feeding boxes, drinking water bottles) in the GLP laboratory barrier environment. 30 minutes immersion in them can achieve more than 99.9% of the sterilization rate. Through the experiments, the best effect can be achieved with the 500mg/l effective chlorine content of 84-disinfectant, 0.1% concentration of benzene of benzalkonium bromide and 1:100 dilution ratio of Baidusha in the sterilization. The effects of these three kinds of disinfectants are positively proportional to the concentration and immersing time.The spraying test results of these three subjects show that 84-disinfectant, benzalkonium bromide, and Baidusha have effective function on the sterilization of walls and air in the GLP barrier environment when the concentration of effective chlorine of the 84-disinfectant reaches 1000mg/l, the concentration of the benzene of benzalkonium bromide is 0.05% and the dilution ratio of Baidusha gets to be 1:600.The experiment of wiping disinfecting method shows that t the sterilization ratio can reach more than 99.9%by using 500mg/l, 1000mg/l and 2000mg/l of 84-disinfectant, 0.05%, 0.1% concentration of benzalkonium bromide and 1:600, 1:200, 1:100 dilution ratio of Baidusha respectively to wipe the surface of the walls and cages every day; if wiping the surfaces of GLP environment barrier walls and cages every two days, the sterilization ratio can reach more than 99.9% with 84-disinfectant chlorine 1000mg/l 2000mg/l; concentration of benzene of 0.1% benzalkonium bromide and dilution ratio for 1:600, 1:200 and 1:100. The dilution ratio of 1:100 Baidusha can maintain 99.9% of the sterilization rate for 5 days. Through the test, the best wiping disinfection effect can be achieved with 1000 mg /l content of active chlorine in 84-disinfectant, 0.1% benzalkonium bromide and 1:100 Baidusha.The results demonstrate that these three studying objects are applicable to the disinfection in barrier environment of GLP laboratory. The immersion sterilization effect is proved positive with 2000mg/l 84-disinfectant concentration of available chlorine concentration, 0.1% benzalkonium bromide and 1:100 dilution ratio Baidusha. The test also proves that the corrosive 84-disinfectant may damage the cages and other stainless steel products, so it’s recommended to be disinfected with benzalkonium bromide and Baidusha. However, the experimental results show that 84-disinfectant is the best choice for immersion sterilization among the three studying objects. If using spraying disinfection once every day, 500mg/L concentration of effective chlorine content of 84-disinfectant, 0.1% benzene tie bromide and dilution proportion to 1:600 Baidusha can reach 99.9% disinfection effect. Wiping is proved reliable for all the studying items, and Baidusha shows the best effect. For the daily wiping sterilization, 500mg/l~2000mg/l 84-disinfectant, 0.05%~0.01% benzalkonium bromide and dilution proportion to 1:600 Baidusha can be the best choice. For sterilization once two days, 1000mg/l~2000mg/l 84-disinfectant, 0.1% benzalkonium bromide and dilution ratio 1:600 Baidusha can help to achieve the best effect. For the sterilization once five days, dilution proportion to 1:100 Baidusha is functional.Considering the disinfection effect, factors of safety, practicality and price synthetically, these three studying subjects are suitable for all kinds of sterilization work in the GLP laboratory barrier environment. For practical disinfecting works, the combination of spaying and wiping are mostly recommended, which shows reliable disinfection effect.
