| Cordyceps Sinensis is a kind of noble Chinese medicine because of it’s extensive pharmacology of herbs and unique living environment. By studing the mycelium of Cordyceps sinensis showed that active ingredients is very similar with natural herbs, the cultured Annmorph of Cordyceps sinensis is a better alternatives of Cordyceps sinensis in the future. In this study, the appeared reason of pink strains of mycelium will be researched, the results as follow:1. A portion of normal fungi strain would changed pink color gradually while cultured cordyceps sinensis fungi. There is a big morphological difference between strain SH-1 and pink strain ZH-1.ITS sequence alignment with normal strains and pink strains shows that the 11 normal strains and 2 pink strains all amplified band, the band is clear and s Tab. the size of every band is 500-750 bp. Using the software BLAST of Genbank analysised similarity and identification, the similarity was 100% between 13 fungi strains and Paecilomyces sp. Cs-4(EU328187) Using Three ways building system evolutionary tree to the ITS sequence of 13 strains and the 32 fungi strains separation from cordyceps sinensis(their ITS sequence downloaded from Genbank). Three methods are shown 13 strains and Paecilomyces hepialid. were divided into a set.It can say 11 normal strains and 2 pink strains all were Paecilomyces hepialid.2.Compared with the different contents of active ingredient in Cordyceps sinensisã€myculim and pick fungi,The result shows that excepting Guanosine and Cordycepin, the rest content of nucleoside is same as normal strains.3. For seeking the reason of emerging pink variation. and obtaining the maximum of yield ratio of the pink fungi. Five main factors including culture time, culture temperature, p H value, loaing volume and sunny condtions, were selected on the basis of experimental results of single factors through using Responce Surface methodology. The optimum culture conditions of the strains SH-1 change to ZH-1 were as follow: culture time was 80 d, culture temperature was 20.55℃, p H value was 7.51, loading volume was 250 m L. Under the conditions, the change probability of ZH-1 was 35.14%; The optimum culture conditions of ZH-1 were as follow: culture time was 80 d, culture temperature was 20.55℃, p H value was 7.51, loading volume was 250 m L. Under the conditions, the change probability of ZH-1 was 35.14%.4. Four main factors including nitrogen A, K2HPO4, nitrogen B, glucose, were selected on the basis of experimental results of single factors through using Responce Surface methodology. The optimum culture condition were as follow: 43.51 m L nitrogen A, 0.6g K2HPO4, 2.56 g nitrogen B, 5.18 g glucose. Under the conditions, the optimum chang probability of ZH-1 was 37.24%。The results showed that the medium formula nitrogen A and K2HPO4 have very important effect on ZH-1 appearing probability, nitrogen A and glucose have very important effect on ZH-1 yield.5. From the above, there was variation of morphology and compositon between normal and pink strains while they were identical species at the molecular level. It suggested that the Morphological diversity and Genetic stability runs simultaneously in this fungi.If there is a connection between the reason and mechanism of shape transform with environmental stress,viability declined, strain aging, degeneration and the invasion of exogenous diseases, the problems need further research.In this paper, the study of new pink strains of Paecilomyces hepialid. laid the groundwork for identification and application. |
