Radix Isatidis, Astragalus Polysaccharide And Interferon On Pig Pseudo Rabies Virus In Vitro Studies

Astragalus polysaccharide(APS) and Isatis root polysaccharide(IRPS) was refine with water and alcohol,determined by anthrone test..In vitro construction of recombinant plasmid p GEX-IFN-α, the recombinant plasmid p GEX-IFN-α transformed into Escherichia coli BL21 and expression of the recombinant protein was induced by IPTG induction.The protein was purified and immunized rat after SDS-PAGE and Western blotting analysis.The inhibitory effect of IRPS,APS and IFN-α on cytopathogenic effect(CPE) induced by PRV in vitro,the direct effect on PRV were observed with the methods of CPE and MTT dye.The inhibitory concentration 50%(IC50) of IRPS, APS and interferon in PRV infected PK-15 cells was 250μg/ml, 500μg/ml and 100.7U / ml. The combined index measuring the drugs in combination, the results show that when the IRPS 125μg/ml, astragalus polysaccharide 250 μ g/ml, and recombinant porcine α-interferon 50 U / m L combination of the three, the best, the virus inhibiting viral suppression effect rate of 70.26%.
