| The distribution of defatted soybean in digestive tracts was studied in presented thesis. This work included three parts: the preparation of defatted soybean nanoparticle( DSN) and the particle characterization; the distribution of defatted soybean(DS) and DNS in digestive tracts; the influence of the digestive tract morphology after DS and DSN administered.Ball mill was used to make the DS into DSN, scanning electron microscopy was used to observe the morphology of DSN and laser particle size analyzer was used to measure the particle size. It was showed that, the particle size of DSN distributed between 200 to 300 nm with good monodispersity. Scanning electron microscope observation showed that, the DSN have spherical morphology, some oval,and some irregular shape. The laser particle size analyzer shows that, the DSN mostly distribution around 200 nm with uniform size, peaked at 219.8nm.Distribution of DSN was studied based on the β-conglycinin’s stability and antigenicity. Double antibody sandwich ELISA was used to quantify the DS, DSN in different parts of the digestive tract. It indicated that, there was no significant difference in distribution between DSN and DS in the digestive tract. Meanwhile they showed the same distributive pattern. Both DS and DSN mainly distributed in the stomach at 0.5h, and after 1h mainly distributed in the small intestine. The similar distributions were observed in DSN group, DS group and fluorescent microspheres group. The three materials distributed in the stomach and the jejunum at 0.5h, then mostly distributed in the jejunum, ileum after 1h, with little in colon. Although in distribution ratio, the DSN group was significant higher than DS group in the colon,the two groups showed no significant difference in the stomach, duodenum, jejunum,ileum, rectum. After lavage 0.5 hã€1 hã€2 hã€3.5 h, the DSN in the digestive tract accounted for 16.8%ã€23.4%ã€24.9%ã€24.6% of the total amount. While the DS in the digestive tract accounted for 17.1%ã€23.6%ã€24.8%ã€24.9% of the total amount.The same time series, the fluorescent microspheres in the digestive tract accounted for17.3%ã€26%ã€30%ã€32% of the total amount.Digestive tract tissue paraffin sections of both DNS group and DS group were stained by HE. The results showed that, the DSN group’s morphological structure of digestive tract was similar to DS group between 0.5h-3.5h. Then colloidal gold dyeing was used on paraffin section. The results showed that, DSN in the digestive tract mainly exists in the cytoplasm of the small intestine plica columnar cells and colon, individual in the stomach of the subcutaneous layer and large intestine plica columnar cells. The individual parts of the colloidal gold appeared aggregation phenomenon. |
