Objective:Animal wool fibers is a indispensable important resource to human production and life,main wool traits are fineness、length and color. The coat color is an important economic traits in wool-producing animal,it is codetermined by many genes.Mouse is a important experiment animals,if we study its coat color formation can contribute to other animal coat color formation.Method:In order to investigate the correlation of Stxl7 gene in mouse hair color formation, this experiment gained the sequence of Stx17 gene,and analysed qualitation and localization of Stx17 gene in different hair color of mouse by Real-time PCR、immunohistochemistry and Western blot technology, so we can further research the mechanism of formation of hair color in mouse.ResuIt:(1)We gained Stx17 gene complete sequence by PCR technology.Compared homologous sequence with other species in NCBI,there is a higher homology with other species. (2)To research the mRNA relative expression quantity of Stx17 gene in different color mouse skin tissue by Real-time PCR,the results showed that mRNA expression level of Stx17 gene in the white and grey skin tissue are 0.115 times and 1.682 times than black skin,the difference is notablely(P<0.01),found gene expression of grey abdomen is significantly higher than white mouse and black back,it showed that Stx17 gene has certain correlation with the formation of grey color. (3)To study the protein expression of STX17 in different color mouse skin tissue by Western blotting,the results showed that STX17 protein expressed in both skin tissue,and the expression level of grey skin tissue is highest(4)The localization of Stx17 gene in mouse skin tissue was tested by immunohistochemistry and the results revealed that it expressed in dermal of mouse skin.And Stx17 gene expressed on membrane of melanocyte by immunochemical staining of mouse melanin cellsConclusion:What is said above showed that Stx17 gene was expressed in both skin tissue,and expression quantity were different. So,we speculate this gene may be associated with the formation of different color.
