| Through the addition of glutamic acid to the basal diet and determination of the parameters about antioxidant capacity in the piglets, we provided more experimental basis for further exploration about the functioning mechanism of the added glutamate on the antioxidant capacity in weaned piglets.By high performance liquid chromatography, intestine slice techniques and real time quantitative PCR, we determined the serum oxidative anzyme vitality, the integrity of intestinal morphology, serum essential amino acids levels and the relative expression of intestinal amino acid transporter gene of weaned piglets under oxidative stress induced by diquat.The results showed that1%Glutamate added in the diet can effectively enhance the organism antioxidant ability of weaned piglets under oxidative stress, enhance the serum T-AOC level (P<0.05); obviously remit the intestinal mucosa damage caused by oxidative stress, remarkably increased the jejunum villus height (P<0.05)and the ratio of villus height and crypt depth in the illum(P<0.05); rise the serum free threonine and methionine levels contrasting with the oxidant group(P<0.05). Besides,1%Glutamate addition can remarkably enhance the intestinal acidic amino acid carrier gene expression (P<0.05) and functionaly downregulate the augmental relative expression of the intestinal alkaline amino acid carrier gene caused by oxidative stress. It follows that glutamate effects the antioxidant performance of piglets under oxidative stress through the intestinal acidic amino acid carrier gene relative expression level regulation, thus effectively relieves the piglets oxidative stress reaction and reduces the damage of reactive oxygen molecules to organism. |
