| Given the probiotics can improve the animal healthy and growth performance and so on that caused the people extensive attention. However, the resistance of probiotics were poorer, the use of microcapsule to embedding probiotics was the research hotspot presenttly. This study on the optimum preparation of calcium alginate microcapsules by emulsification, using the optimization technology embedding Saccharomyces boulardii and Enterococcus faecium, research on their growth conditions, and the stress resistance in the simulated condition of high temperature and high humidity, gastrointestinal juice. Finally, the microencapsulated organisms were further applied to broilers for the confirmation of the application effect. The results shows that:(1) The optimized conditions for micro gel beads preparation was:2g/L of the surfactant Span80,12g/L of the alginate,1:4of the mass ratio of CaCO3to NaAlg and1:3of the volume ratio of the water to oil. In addition, Experiments further showed that after repeated use of the liquid paraffin for ten times, micro gel beads still can keep a good morphology and a higher yield.(2) Microencapsulated Saccharomyces boulardii and Enterococcus faeciumin not noly have good growth characteristics, its resistance also significantly improve. The microencapsulated Saccharomyces boulardii and Enterococcus faeciumin can have about25%and40%increase respectively in survived rate than free ones under high temperature and humidity conditions,60%and25%were increasing for microencapsulated Saccharomyces boulardii and Enterococcus faecium respectively than free ones in simulated gastric juice condition. The data were15%and20%when the simulated intestinal juice was concerned.(3) Contrast to the non-capsulated Saccharomyces boulardii, the broilers feed contained microencapsulated Saccharomyces boulardii (10cfu microencapsulated Saccharomyces boulardii/kg daily diet) can improve the growth performance trend, also can increase the index of spleen and bursa of fabricius, and improve the immune performance of broilers to some extent.Above all,preparation of pre-fermented microencapsulated probiotics by optimized technology, the proliferative density of probiotics in capsule were significantly improved compare probiotics of non-capsule, also can significantly improve the ability to resist high temperature and high humidity, simulated gastric intestinal juice. |
