| In this study, screening the SNP of Keratin genes(KRT26, KAP16)and FAIM genes by using DNA mixed pool re-sequencing and MassArray technology; Chinese merino(Xinjiang type)sheep as experimental materials, the FAIM, KRT26 and KAP16 genes were conducted genetic analysis,the results are following:1. Applied DNA mixed pool resequencing technology to conduct gene sequence analysi s of KRT26, KAP16 and FAIM genes and were detected 29 SNPs, in which 16 were syno nymous mutations,13 were non-synonymous mutations.FAIM gene have two exons and two non-synonymous mutations.KRT26 gene have four exons, two non-synonymous mutations an d six synonymous mutations.KAP16 gene have four exons,nine non-synonymous mutations a nd 10 synonymous mutations. In accordance with the requirements of typing technology, fin ally screened six useful SNPs, such as C248582270Tã€C40544613Gã€C41006938Tã€A41009186Gã€T41006710Cã€A41007121C, respectively.The C248582270 T mutation point exchanged the 83 Amino acids of exon 2 from Glycine(Gly) into Glutamate(Glu); the C40544613 G a nd A41009186 G mutation points exchanged the 59-Amino acid and 69-Amino acid in exon 1from Glycine(Gly) into Arginine(Arg) and Valine(Val) into Alanine(Ala), respectively;the C41006938 T, T41006710 C and A41007121 C mutation points exchanged the 375-Amino acid, 1451-Amino acid and 314-Amino acid in exon 5 from Glycine(Gly) into Serine(Ser),Isoleucine(Ile) into Valine(Val) and Cysteine(Cys) into Glycine(Gly), respectively.2. Using MassArray technology in order to typing for the SNPs of KRT26, KAP16 and FAIMgene; analysis the relationship between genotype and wool traits by using SAS8.1 sof tware, the re-sults are following: C41006938 T point have a very significant influence on wei ght after shearing. A41009186 G point have a significant influence on bending number. T41006710 C point have a significant influence on fiber distribution, variation coefficient and iden tification of weight. In C41006938 T point in KAP16 gene, the weight after shearing of CC genotype was significantly lower than CT and TT genotypes; in A41009186 G point, the ben ding number of AA genotype was significantly higher than GA and GG genotypes; in A41006710 C point, the fiber distribution, variation coefficient and identification of weight of CC genotype were significantly higher than CT and TT genotypes.3. Six SNP were conducted linkage disequilibrium analysis by using Haploview version4.1 software, in which 4 SNP to make up a haplotype Block, they are C41006938Tã€A41009186Gã€T41006710Cã€A41007121C respectively. T41006710 C and C41006938 T was in stro ng linkage disequilibrium state; T41006710 C and A41009186Gã€C41006938T and A41009186 G were in linkage disequilibrium state; the variation coefficient and weight after shearing o f Haplotype in C41006938 T and A41009186 G points were significant differences than other haplotypes. |
