The main material of this research were52cotton caltivars (lines) and M37(♂Xinhai20×♀Shuofeng1),M44(♂Junmian1×♀Liaomian18), series4population(♂Junhai1×♀Liaomian18) and10Q population(♂Xinhai16×♀108Fu). Acupuncture injection of stem and a toothpick inserted by inoculation, to observethe incidence of various Physiological period, and to investigate the yield traits of agronomic traitsin harvest period;dynamic SOD、POD、CAT activity and MDA content in different resistance of G.barbadence and G.hirsutum cultivars were measured in seedling leaves inoculated by Verticillium dahliaeafter symptomatic in the room. Through research of the relationships between changes of superoxidedismutase (SOD), peroxidase (POD), catalase (CAT) and malonaldehyde(MDA) and resistance of cotton toVerticillium wilt, povide the basis for the identification of resistance. In this study, screening34pairs ofSSR primer for hybrid M37groups and hybrid M44groups of male, female parent of andresistant, susceptible gene pool. Amplified polymorphism in different bands, markers were detectedin M37and M44population. To analyze the relationship between markers and the relative disease index, inorder to improve the efficiency of marker assisted selection, which can provide a reference markerfor disease resistance breeding.(1) In different resistance to Verticillium Wilt of cotton caltivars (lines) and M37groups, M44groups,4series of groups and10Q series groups, relative disease index and lint percentage was negativelyrelated and significant negative correlation relationship. In upland cotton, M44groups,4series of groupsand10Q series groups,relative disease index and fibre weight per boll was significantlynegativecorrelationand a very significant negative correlation relationship. In addition, the10Q series groups, relative diseaseindex and boll weight was negatively correlated, therefore, lint percentage, un-ginned fiber weight and bollweight can be used as three factors to measure the yield traits.(2)With the induction of V.dahliae,SOD activity decreased,SOD activity in the leaves of cotton thanbacteria was dropped,Which disease-resistant, susceptible caltivars (In>20) is greater than the decrease inthe amplitude of high resistance,resistant caltivars (0<In≤20). While, MDA levels than non-inoculatedrise, especially resistance to disease, susceptible caltivars (In>20) is much larger than the increase inhighly resistant,resistant caltivars (0<In≤20). With the induction of V. dahlia, Cotton seedling leavesdifferences POD, CAT activity than the activity generated by the difference Missed bacteriasignificantly.Using induction of V. dahliae, the measure effect of physiological and biochemical product isbetter than not inoculation of cotton. So, SOD activity and MDA content can be used as identification ofresistance to Verticillium wilt of cotton physiological and biochemical parameters. Product is better than notinoculation of cotton.(3) In the F2:8group as the material, by using34pairs of primer SSR polymorphism on resistant maleparent, susceptible female parent, resistant gene pool, susceptible gene pool, interspecific hybrid of M37generation and M44generation, BNL1414, BNL1721and JESPR114could amplified polymorphism indifferent bands. Mark BNL1721and the relative disease index had significantl correlation inthe hybrid group M37, no significant hybridization in one group, which may have relations with the islandcotton Verticillium wilt resistance gene itself. Thus, Marker BNL1414amplified differential bands in theparental Xinhai20and Shuofeng1of M37group, while, no amplified differential bands in the parentalLiaomian18and Junmian1. Marker BNL1414had significant postitive correlation with the relative diseaseindex of M37group, so marker BNL1414can do for identification of sea island cotton, uplandcotton, but also can be used as evaluation index of resistance to Verticillium Wilt of molecular markers.Marker BNL1721amplified differential bands in the group M37and M44. But, the relative diseaseindex was significantly associated markers BNL1721and group M37, no significant correlation withthe relative disease index of group M44.So, SSR polymorphic primers BNL1414and BNL1721not only can be used as a marker forscreening cotton caltivars, resiseant and susceptible gene pool, and can be markers for identificationof sea island-upland hybrid generation. As can be used to compare the results of QTLpositioning of cotton Verticillium wilt resistance is very few, so the genetic marker test results have yet tobe further analysis and research, especially the analysis of a large number of molecular marker of QTL.Therefore the mark to be further in-depth study.
