Polygonum perfoliatum L., which belongs to Polygonum plants, has anti-inflammatory, antivirus, antitumor, cough-relieving, pests-killing and other active functions. This trail employed Tenebrio molitors for test and selected anesthetic activity as well as the growth factors like the weight change and the molting of Tenebrio molitors as screening index, providing a systematic study for the active ingredients in Polygonum perfoliatum L. to develop its medicinal value. The results showed:(1)Anesthesia effect of extracts from different layers of Polygonum perfoliatum L. on Tenebrio molitors of different ages in which the 3 instar larvae was the most sensitive had dose-dependent effect, which was remarkable within 10 min of anesthesia time.(2)Petroleum ether extracts had the best anesthesia activity, especially with the concentration of 4mg/ml on 3, 8 age of Tenebrio molitor. The anesthesia correction rate was 100% after 3min. LC50 was 0.0083mg/ml and 0.0589mg/ml respectively, lower than other extracts. The anesthetic activity of n-butanol extracts was next, of which LC50 was 0.4524mg/ml after 3min on 8 age of Tenebrio molitor.(3)The extracts of ethyl acetate had the most obvious effect on weight of 3 age of Tenebrio molitor in 16 days, in which the weight growth rate was lower than the control group compared with others. But there was no significant difference in different concentrations, which also needs further verification.(4)3, 6 and 8 age of Tenebrio molitor disposed by the extracts of Polygonum perfoliatum, completed the first molt on 8, 6, 10 days respectively. The petroleum ether extract of different concentrations had significant difference from the control group on the molting of 6 age of Tenebrio molitor. But there was no significant difference in different concentrations, which also needs further verification.Therefore, the anesthesia activity components locate in the smaller polarity range of petroleum ether mainly and the bigger one of n-butanol. The active compounds influencing or inhibiting the growth of weight were the ethyl acetate extracts to which the weight of young Tenebrio molitor were more sensitive. While the active substances influencing molt and growth of Tenebrio molitor were the petroleum ether extracts, and the molting was also related to the age of Tenebrio molitor which means the older it gets, the later the first molt time is longer. Thus the material and age were both important factors to affect the changes of weight and molting. To combine the material and activity, specific active ingredients need further isolation and identification so as to develop botanical pesticides.The HPLC-ESI-MS was employed for isolation and identifition of Polygonum perfoliatum with petroleum ether, ethyl acetate and n-butanol. Through the natural compound library web site(http://kanaya.naist.jp/knapsack_jsp/top.html) and other related references, 58 compounds were identified from Polygonum perfoliatum L. in which 47 were the first time to be identified including 25 kinds of fat acid(compound 4、11~34), 7 flavonoids(compound 35~41), 3 phenylpropanoids(compound 5、42、50), 4 steroids(compound 6、51~53), 3 terpenes(compound 7、8、54) and 5 other types(compound 10、55~58).
