| In this experiment, the optimum addition amount of Allium mongilicum regel flavonoids in basal diet for sheep was filtered according to the effects of Allium mongilicum regel flavonoids on ruminal fermentation parameter in vitro. And then we studied the effects of Allium mongilicum regel flavonoids on rumen inner environment indices and fiber degradation, so that this study could further provide scientific basis for applying intensive processing products of Allium mongilicum regel flavonoids into meat goat feeding. The dissertation included three experiments following:(1) Four ruminally fistulated Mongolian male sheep (body weight,34.6±1.5 kg) were used in a single factor experimental design and batch culture of artificial rumen in vitro. In this study, we added 0.000 (control),0.022,0.032,0.042,0.052 mg of Allium mongilicum regel flavonoids in every 2g of substrates in order to filter the optimum addition amount of Allium mongilicum regel flavonoids in basal diet. Results show that:1) Allium mongilicum regel flavonoids supplementation in basal diet increased gas production significantly, and 0.032mg/2g increased the biggest (P<0.05); 2) Compared with control treatment, Allium mongilicum regel flavonoids supplementations increased the pH of culture solution numerically, and 0.032mg/2g and 0.042mg/2g significantly increased the concentration of BCP, and decreased NH3-N concentration of culture solution significantly (P<0.05); 3) Allium mongilicum regel flavonoids supplementations improved TVFA, but markedly decreased the ratio of acetate/propionic acid (P<0.05). Therefore, the most optimum addition amount of allium flavonoids for sheep fed basal diet with concentrate to forage 3:7 was 0.032~0.042 mg/2g, that was 16~21 mg/kg.(2) According to the self-control design of experiment, the treatments were:1) control; 2) Allium mongilicum regel flavonoids supplementation treatment 1,16mg/kg; 3) Allium mongilicum regel flavonoids supplementation treatment 2,21 mg/kg. Each period lasted for 15 day, consisted of 12-day preliminary period and 3-day experimental period. Rrumen fluid samples were collected for ruminal environmental parameter analysis at 0 h before feedind and 2 h,4 h,6 h,8 h,10 h,14 h,18 h and 22 h after feeding. Results demonstrate that:1) Diet supplemented with Allium mongilicum regel flavonoids increased pH of rumen fluid numerically without significant difference between treatments; 2) Diet supplemented with Allium mongilicum regel flavonoids decreased NH3-N concentration of rumen fluid; 3) Allium mongilicum regel flavonoids supplementation increased bacterial protein concentration (P<0.05); 4) The total VFA concentration and propionic acid concentration were increased significantly, but acetate/propionic acid was decreased (P<0.05).(3) Four ruminally fistulated Mongolian male sheep were used in the self-control design of experiment by using nylon bag technique. The experiment consisted of two periods with 19 days each period, including 14-day preliminary period and 5-day experimental period. During the experimental period, nylon bags were placed at rumen saccusventralis at Oh,2h,6h,24h,36h,48h and 72h, which were used for analysing degradation rate of cellulose. Results show that:1) Compared with control treatment, degradation rate of DM and NDF tended to be increased by Allium mongilicum regel fiavonoids supplementation at different time points, and the difference among supplementation treatments was significant (P<0.05); 2) Allium mongilicum regel fiavonoids influenced the ruminal kinetic parameters, particulaily, the supplementation treatments increased rapid degradability a of DM relative to control treatment, and the supplementation treatments increased slow degradability b of DM numerically; 3) The effective degradation rate of DM was increased significantly for Allium mongilicum regel flavonoids supplementation treatments compared with control treatment. |
