Preparation And Physicochemical Properties Detection Of Sheep Spleen Transfer Factor And Its Impact On Immune Function In Mice

Object:This experiment is based on transfer factor (TF) which raw material was sheep spleen, the preparation method of sheep transfer factor(S-TF) is explored, and the preparation principles and methods are defined.First, S-TF is crudely extracted to analyze its physicochemical property preliminarily, then animal test is done to exploring that if S-TF can danmage anamal’s organs microcopically and that if it have the function to improve immunity.Methods:.This research was based on many literatures and S-TF was taken by the method of thaw-dialysis. After trial and error, the preparation steps are ensured, and S-TF is taken, physicochemical properties are analyzed, experimental animal safty evaluation and the influence of immune function in mice is explored. Analysis of physicochemical properties including that the ratio of appearance, pH value, protein reaction,polypeptide content determination, Determinnation of ribose content, the ratio of nucleotide and polypeptide (OD260/OD280), Determination of Amino acid composition and content, mineral element contents, sterility testing. Animal experiment includes two parts.The first part is that the mice is drenched with crudely extracted S-TF in 15days, taken blood through eyeball to do the Blood routine test, then spleen index is determined and heart, lung, liver, spleen, kidney, stomach, duodenum, jejunum, ileum, cecum, colon, rectum are taken to make pathological section and microstructure examination; the second part is that phagocytosis of chicken red blood cell test in mouse peritoneal macrophages is done in 19 days and the phagocytic percentage and index is determined. Through the two parts experiments, whether S-TF can improve the immune function is tested.Results:The basic principle and method of preparing are mastered though trial and error to making S-TF. Though analyzing the physicochemical properties, all results are complined with the national standard of TF.Animal experiment results show that the number of white blood cells of experimental mice, especially the number of neutrophils was significantly increased; the organizational structure of some orgins in mice without obvious pathological injury by light microscope; there are no significant differences in the spleen index; S-TF can increase the phagocytic percentage and phagocytic index of peritoneal macrophages in mice, indicating that S-TF has the function to increase the immune of mice.
