Background:Transgelin2(Tagln2), known as SM22β, belongs to Calponin family and is involved in cell proliferation and differentiation of animal development. Tagln2 is an actin-binding protein, through affecting microfilament polymerization to regulate microfilament skeleton,Further involved in the regulation of cellular functions, for example:cell proliferation, differentiation, migration and apoptosis. Our previous studies indicated that Tagln2 contributes to embryo implantation in mice and rabbits. Whether Tagln2 play a role in goat uterus during the early pregnancy,it’s worth further study.Methods:The expression of Tagln2 was examined on Days 0,9,12,and 16 during the estrous cycle and Days 6,9,12,15,22,25,30,40,50, and 60 during the pregnancy of goat uterus using In situ hybridization and Western blot. And after two months leter of ovariectomy,use estrogen, progesterone, estrogen and progesterone common treatment, oil treatment (control) with goat.Tagln2 were also detected by the same methods above in ovariectomized goats were treated with progesterone and estrogen.Results:Tagln2 mRNA was mainly expressed in luminal epithelium and glandular epithelium in uterus of during the estrous cycle.Tagln2 mRNA was detected in uterus of during the estrous cycle, the signal stronger at day 9 of estrus. Results of Western blot showed that Tagln2 protein was detected in uterus and peak in day 9 of during the estrous cycle.Tagln2 mRNA was also mainly expressed in luminal epithelium and glandular epithelium in uterus of during early pregnancy.The level of Tagln2 mRNA increased as pregnancy processed and become stronger after 25 days of pregnancy, the adhesion sites was significantly higher than that of non-adhesion sites. Expression of Tagln2 protein was consistent to the mRNA,and significantly up-regulated during early pregnancy.In the steriod hormonal treatments, the results of in situ hybridization and Western blot were showed,that the expression of Tagln2 up-regulated by P4 treatment,but there were no significant change in group by E treated.Conclusion:Our results of In Situ hybridization indicated that expression of Tagln2 showed embryo implantation specificity in luminal and glandular epithelium of goat uterus during implantation and becomes stronger as the process of implantation and placentation. We speculate that Tagln2 participates in implantation and placentation during early pregnancy in goats. P4 up regulated the expression of Tagln2 and E2 had no significant effect on it.
