Bovine papillomavirus(Bovine Papillomavirus BPV) is pathogen of bovine papilloma. BPV can cause bovine skin and epithelial hyperplasia. BPV has been aglobal epidemic trend for now, which brings huge economic losses in cattle industry. The methods to prevent and treat bovine papilloma are surgical treatment method, traditional Chinese medicinetherapy and autoimmunity treatment method. This study used BPV to infect C127 cells, established cell infection model and screen treatment drugs in vitro.The experiment selected clinical typical bovine papilloma tissue, observed it by OM and TEM, detected BPV in tumor tissue by indirect ELISA method, determined the genotype though PCR amplification and sequencing. We used wild strain BPV to infected C127 cells, in contrast to cell morphology changes, and observed virus particles distribution using TEM. Taking Leigh Bhave Lin as a positive control, we detected matrine and Oxymatrine direct inactivation ability, inhibiting virus proliferation ability in cells and preventing virus infection ability on BPV. Taking BPV wild strain to infect C127 cell model as platform, we evaluated effect of matrine and oxymatrine on BPV.Conclusion: We could see multiple finger-like protrusions, the central of each projection was fibrous tissue and blood vessels, the outside of protruding was multilayered squamous epithelium cells though OM; little atypia, no significant keratosis and no abnormal mitotic. Moreover, we could see basal cells, prickle cells and granule cells presented pathological changes in irregular cell shape and vacuolated mitochondria though TEM. BPV tumor tissue was detected with indirect ELISA showed positive reaction. And gene similarity between tumor virus DNA sequencing and separation from China SW BPV-2 virus strains(Gen Bank: KC256805.1) L1 was up to 99%. C127 cells, infected by BPV in vitro, becomed small、elongated, growth-intensive, disorganized, cell overlap to form a cross. PCR detection appeared specific bands at 450 bp. By TEM we found virus particles and pathological changes like Golgi apparatus, rough endoplasmic reticulum, mitochondria deformation. Conclusion: papillomas on the surface of calves were caused by BPV-2, and BPV-2 could infect C127 cells which showed morphological changes.Conclusion: Matrine can directly act on BPV to make the activity lose, it inhibited the proliferation of BPV in the cell, and prevent BPV infection.
