Pregnancy is one of the most important issues in dairy industry. The expression of genes which related to pregnancy plays a vital role except the nutrition and management in dairy cows pregnancy. It has been reported that the uterus produces local hypoxia microenvironment before the establishment of embryo implantation. Our previous study also showed that hypoxia can affect the expression Bo LA-A, which is an important gene for the bovine matrix to recognize the embryos.MUC1 expresses in ovarian, breast, pancreas, uterus, other epithelial tissues and organs. It is a high molecular weight, highly glycosylated transmembrane protein, which plays an important role in cell signal transduction, cell adhesion regulation. In addition, it can also lubricate and protect epithelial tissues. Previous studies have shown that the highest expression during the implantation window was in the mid secretory stage in endometrial epithelia in human. The expression of MUC1 is correlated with the implantation rate, therefore, it was used as one of the endometrial receptivity markers.S100A11 belongs to calcium binding protein S100 family, which mainly expresses in kidney, lung, skin and other various organizations in placenta. It is a messenger of cell signal transduction by binding to calcium ion, and it is related with cell cycle, apoptosis, inflammatory diseases and cancer. Previous studies demonstrated that the low expression of S100A11 in human endometrium during the window period affected the implantation and abortion with some unknown reason.Previous studies showed that both of the two genes played crucial roles during the window of implantation, and this period was anoxic on the maternal-fetal interface. The present study mainly explored the expression of MUC1 and S100A11 in endometrial epithelial cells(EEC), and further analyzed the effect of hypoxia on their expression in EEC.We established the hypoxia model with Co Cl2 for EEC as previous published report. The expression of MUC1 and S100A11 were analyzed with RT-PCR and Western blot. The results showed that: MUC1 and S100A11 expressed in EEC, and the expression were regulated by hypoxia in EEC treated with different concentrations of Co CL2(100 umol/L, 200 umol/L and 400 umol/L). MUC1 expression was up-regulated in EEC treated with 200 umol/L and 400 umol/L Co CL2 at 3h at m RNA level, and all doses of Co CL2 increased its expression at 6h and 12 h. In addtion, 400 umol/L Co CL2 significantly affected its expression than the other groups. All the different concentration of Co CL2 affected S100A11 expression at m RNA level, and 400umol/L Co Cl2 had the strongest effect at 12 h. Western blot results showed that only MUC1 was up-regulated in 400umol/L group at 12 h, the other groups could not increase MUC1 and S100A11 expression in ECC. Together, the results showed that MUC1 and S100A11 were increased significantly in hypoxia EEC treated with Co Cl2 at m RNA level, and only MUC1 significantly increased treated with 400umol/L Co Cl2 at 12 h at protein level. We speculated that MUC1 might participate in the uterus inner environment construction of dairy cows during embryo implantation.
