Pinellia ternata(Thunb.) Breit is a traditional rare medicinal herbs, which drying tubers could be used for medicine, belongs to Araceae Pinellia ternata species. Its medication history had been more than 2,000 years. However, as for excessive excavation, wild Pinellia ternata has became increasingly depleted. At the same time, cultivated Pinellia ternata are affected by various viruses and pests seriously, with the result that yield decreasing year by year, quality deteriorate seriously, which is unable to satisfy the pharmaceutical market for raw materials normalization and standardization requirements.In this paper, we chose Pinellia ternata shoot tips as the explant, utilizing the method of tissue culture to obtain breeding of virus-free seedling. Apart from this, we also studied the following research areas in one step to establishment of seedling technology and standardization cultivation techniques of Pinellia ternata:1. Breeding of virus-free culture seedling of pinellia ternata: Using Pinellia shoot tips as explants, using two factors and three levels orthogonal experiment method to screen out the most suitable medium for inducing callus: MS+2.0mg/L2,4-D+1.0mg/L KT+30g/L sucrose+7.5g/L agar powder, getting a pale green granular callus growwing well. The callus subculture proliferation of two times adapter in clustered buds media: MS+1.0mg/L2.4-D+1.0mg/L6-BA+1.0mg/LNAA+30g/L sucrose +7.5g/L agar powder. Buds differentiation into the height of about 3cm plantlets and transfer to the rooting medium, selecting the best rooting medium: MS+0.5mg/L IBA+0.2mg/LNAA+30g/L sucrose +7.5g/L agar powder, the plant growth well, root robust, transplant survival rate of 90%, detoxification rate of 88%.2. One step to establishment of seedling: Pinellia tuber as explants, the optimum time is that after alcohol disinfection sterilization 15 s and then HgCl2 disinfection 15 min to screen out the best culture medium of one step to establishment of seedling of Pinellia. The best culture medium is MS+1.0mg/L6-BA+0.5mg/LNAA+30g/L sucrose+ 7.5g/L agar powder, under this condition, transplant survival rate attains 85%.3. Standardization cultivation techniques of Pinellia ternata: Pinellia ternata per plant yield has a closely positive correlated with main leaf length, plant height, leaf width and the main stem diameter. In the high-yield cultivation technology of Pinellia, controlled by single factor, we set three different kinds of stem size, five kinds of planting density, five harvests.We conclude that Pinellia’s production is significantly affected by the kinds of stem size, density of planting and the harvest period.When the seed stem size is 1.0-1.5cm, harvest stem tubers andpropagation coefficient are the highest. In the 10 cm × 12 cm planting density, the harvest index and propagation coefficient are the highest. The tuber yield is the highest when harvest in late August and early September.
