In recent years, increasingly people paid more and more attention to the food contamination. The residues of veterinary drugs is one of the representative part. Veterinary drugs is a very important part in aquaculture and animal husbandry. But the unreasonable, indiscriminate use of veterinary drugs will harm the human health and affect international trade. Animal original food are mostly complex matrix, interference, a simple and effective purification method is needed. For the determination of trace residues of veterinary drugs, SPE is a nice method, it can extracted trace target compounds from the complex matrix, and can enrichment and concentration. We establish a sensitive and selective liquid chromatography-tandem mass spectrometry method for the simultaneous identi?cation and quantitation of polyether antibiotics in animal original food. This paper includes three parts as follows:In Chapter one, the background and the significance of this dissertation was elucidated. Introduce the principle and development of SPE method and on-line purification pretreatment method and basic theory of MS and MS/MS.In Chapter two, a sensitive liquid chromatography-tandem mass spectrometry(LC-MS/MS) method was developed for the simultaneous determination of polyether antibiotics in animal original food. Five kinds of SPE cartridges were applied by comparing the matrix application, matrix effect and maneuverability and we use carbon black solid phase extraction column as purification column. This method is sensitive, reproducible and suitable for the determination of polyether antibiotics in milk, chiken muscles, eggs and chiken livers.In Chapter three, a new method using Turbo Flow on-line cleanup liquid chromatography-tandem mass spectrometry has been developed for simultaneous determination of six polyether antibiotics. This proposed method is simple, rapid, sensitive and highly selective, and can be applied to simultaneous identification and quantification of the six polyether antibiotics in animal original food.
