Corydalis yanhusuo W. T. Wang, a herb belongding papaveraceae Corydalis D C., fromdifferent origins, was used as the research material. The main contents of research work forCorydalis yanhusuo W. T. Wang was divided into six parts, which respectively were thefollowing, i) the quality standard specification of Corydalis yanhusuo W. T. Wang, ii) toanalysis and detect alkaloid content of Corydalis yanhusuo in different regions, iii) themapping fingerprints of Corydalis yanhusuo in different regions by High Performance LiquidChromatograpHy, iv) determination of the trace elements by flame atomic absorption,v)determination of heavy metal by graphite furnace atomic absorption, vi) analysis oforganochlorine and pyrethroid pesticide residues by gas chromatography-mass spectrometry.The six parts of the main research content in the order:1. Research on quality standards by The Chinese pharmacopoeia. The moisture, ashcontent and the content of alcohol extract of Corydalis yanhusuo from different origins wasdetermined by the2010edition of China pharmacopoeia, according to the provisions ofpharmacopoeia, the moisture content exceeded1.5%in some samples; The total ash contentcan’t exceed4%, acid insoluble ash content can’t exceed1.5%, the ash contentdetermination results can be seen that the test sample is in conformity with the requirements,RSD is within5%; The determination of alcohol soluble extract RSD within2%, the contentconform to the requirements of the pharmacopoeia of China shall not be less than13.0%.2. The determination on five kinds of alkaloid content of Corydalis yanhusuo fromdifferent regions. To analysis and detect content of5kinds of alkaloids in Corydalis yanhusuoherbs from13different origins, and alkaloid content differences of the typical medicinalmaterials with the other medicinal are compared. Methods: using ultrasonic assistedextraction method to extract the sample, combined with3D map qualitative identification,selection of HPLC quantitative standard method, including the dl-tet-rahydropal-matine,coryaline and protopine are a good linear relationship between0.05and0.5μg/μL, so aretetrahydroberine and dehydroglaucine in0.1008and0.01008μg/μL, regarding to regression equation, R2is more than0.999. Results: thedl-tet-rahydr opalmatine, coryaline, pro-topine,tetrahydroberine and dehydroglaucine, which average sample recovery rate was between84.6%and96.2%, the5kinds of alkaloid content of medicinal materials from differentregions was different, the alkaloids content of medicinal materials from same regions alsoappear large difference, but the ones of different regions existed the same proportion ofalkaloids. Conclusion: compared to the alkaloid composition, origin of Corydalis yanhusuoplay an important role in its alkaloids content.3. Research on HPLC fingerprint. To establish13kinds of Corydalis yanhusuo fromdifferent regions by HPLC-3D fingerprinting, and comparing with different sources offingerprint of corydalis. Methods:5kinds of main alkaloids as indicator, based ontwo-dimensional map of single wavelength analysis and construction of3D fingerprint, thesimilarity evaluation and the peak of the analysis of their differences. Results: the total12peaks, defining three characteristic peaks in3D fingerprint chromatogram, all of similaritywas in90%above, the cluster of non-public peak analysis showed that different groups ofdifferent sources of Corydalis yanhusuo. Conclusion: the3D fingerprint of Chinese herbalmedicine ingredients can reflect more comprehensive, has important significance in thequality control.4. Study on trace elements. By flame atomic absorption spectrometry, respectively inCorydalis yanhusuo five kinds of trace elements (Fe, Zn, Cu, Mn, Ni) content was analyzed,and the instrument precision was precise, the recovery was96.41%~96.41%, the content oftrace elements have a certain amount of differences for different regions, but The highestlevels of metal elements was Fe, followed by Mn, Zn, Cu, Ni, this research, to a great extent,effectively reflect the changes on the origin of trace elements content, and provide scientificbasis for quality detection.5. Research on heavy metal content. Heavy metal content in Corydalis yanhusuo ofdifferent regions was analyzed with graphite furnace atomic absorption spectrometry, and theinstrument precision was precise, the recovery was95.5%~95.5%, the determination ofmercury (Hg) and arsenic (As) is less than2.0mg/kg, mercuryis less than0.2mg/kg, ministryof foreign trade and economic cooperation conforms to the requirement "green trade standardsof importing medicinal plants&preprations" requirements. 6. Analysis on organochlorine and pyrethroid pesticide residues. Establish18kinds oforganochlorine and pyrethroid pesticide residues of a simple and rapid capillary gaschromatography mass spectrometry analysis method in Corydalis yanhusuo, to inspectwhether accord with standard of green traditional Chinese medicine, provide a reference forthe quality safety. Study on comparing two methods about extraction and purification oforganochlorine and pyrethroid pesticide. Extraction and purification liquid the DB-17capillary column, gas chromatography mass spectrometry (GC-MS) detection, qualitative ionwas choose to quality pesticide, by external standard method determination of pesticide.Results: some Corydalis yanhusuo samples detection might be pyrethroids, and its content arenot beyond the standard. Conclusion: the samples of Corydalis yanhusuo are accord withstandard of green traditional Chinese medicine.
