Study On The Pharmacodynamics Of The Rufangyan Rehabilitation Solution

Bovine mastitis is a complex, common and frequent disease, which affects lactationdisorder and causes serious economic losses to dairy farm. It is difficult to prevent and controlbecause of the pathogens and the complicated pathogenesis. According to the principle ofsyndrome differentiation and treatment of traditional Chinese veterinary medicine and thepathological features of mastitis, in combination with modern new process and newtechnology, the Chinese herbal compound Rufangyan rehabilitation solution(Ruyankang)liniment who was transdermal absorption was developed, which was composed of LoniceraJaponica, Herba Taraxaci, Viola philippica Car, Sophora flavescens, and penetrationenhancer azone, bornel, and so on. It had the effects of heat-clearing and detoxifying,antisepsis and anti-inflammation, promoting milk flow and dredging the meridianpassage, decreasing swelling to alleviating pain.Objective: In order to study the efficacy of “Ruyankang”, the in vitro bacteria inhibition,anti-inflammatory test, skin irritancy test, acute toxicity test, and clinical treatment test werediscussed to provide theoretical basis for clinical application.Methods:(1) Through the tube double dilution method, detected the“Ruyankang”minimal inhibitory concentration(MIC) and minimal bactericidalconcentration(MBC) against the main pathogenic bacteria(Staphylococcus aureus,Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis and Escherichiacoli);(2) the auricle swelling in mice induced by xylene, the mice granuloma induced byplanting cotton ball and capillary permeability raised by acetic acid in mice were used tostudy on the anti-inflammatory activity;(3) the oral acute toxicity of mice (LD50), the skinacute toxicity of rabbit, and rabbit skin irritation experiment were used to test the safety of“Ruyankang”.(4) on the basis of bacteria inhibition, anti-inflammatory and safety test,36dairy cows with clinical mastitis were selected and divided into “Ruyankang” high, mediumand low group, and Compound Honeysuckle injection group,the clinical treatment test wastook, and the milk pH, the aspartate aminotransferase(GOT), alanine aminotransferare(GPT),lactate dehydrogenase(LDH), alkaline phosphatase(LKP) were measured to investigate theeffect of prevention and control. Results:(1) In vitro test, it indicated that the “Ruyankang” MIC against the five kindsof main pathogenic bacteria, were31.25mg/mL,15.63mg/mL,32.25mg/mL,62.5mg/mL,62.5mg/mL, respectively, and the MBC were62.5mg/mL,32.25mg/mL,62.5mg/mL,125mg/mL,125mg/mL, respectively, so it showed the depressant effect to the five bacteria.(2)The anti-inflammation test revealed that the “Ruyankang” medium dose could significantlyinhibit ear-swelling in mice induced by xylene, significantly inhibit the mice granulomainduced by planting cotton ball and capillary permeability raised by acetic acid in mice. Theresult indicated that “Ruyankang” had a satisfactory antagonistic effect on active and chronicinflammation.(3) First, the back skin of rabbits were denuded and smeared with “Ruyankang”at high (4g/mL) and low dose (0.23g/mL) after one side was damaged, and rats were pouredinto stomach. The acute toxicity reactions in the skin and whole body were observed for sevendays. The results showed that there were no any abnormal reactions. Through the oral acutetoxicity test that the LD50was44005mg/kg indicated it had no toxicity. the back skin ofrabbits was smeared with the “Ruyankang”. and then the stimulation and acute toxicity to skinof the drug were observed. The results indicated that there were no erythema and edemaexcept for the damaged skin in one rabbit. Those suggested that the “Ruyankang” were safety.(4) Clinical experiment shows that the effective rate of clinical mastitis by "Ruyankang" high,medium, low group and compound honeysuckle injection were respectively84.21%,85%,68.42%,66.67%, and the cure rate were respectively63.16%,65%,52.63%,52.38%, and“Ruyankang” has a significant treatment effect, the cure rate and effective rate is significantlyhigher than compound honeysuckle injection, and could obviously reduce the milk pH, GOT,GPT and LDH and AKP activity.Conclusion: The bacteriostasis, and the anti-inflammation activity of “Ruyankang” wasremarkable, and it belongs to the non-toxic, no stimulation transdermal absorption drug.It hada good effect in clinical prevention and cure.
