Peste des petits ruminants(PPR) is A class animal infectious disease notified by the OIE. It had outbroken for many times in China since2007and was severely harmful to the health of small ruminants such as goat and sheep, which caused a huge economic losses.There are some diagnostic methods for the detection of the peste des petits ruminants virus based on NP protein, but the information of linear B cell epitopes about the N protein is not detailed and complete. In this study, the16or21mer peptide gene fragments (from P1to P64) covering N protein and the corresponding8-15mer peptide gene fragments were synthesized using the gene biosynthesis strategy according to the full gene of PPRV-China/Tibet/Geg/07-30. The synthetic gene fragments were cloned into pXXGST plasmid and the prokaryotic expression vectors were constructed, and then the recombinant proteins with GST were obtained. The distribution of linear B cell epitopes (BCE) of N protein and the corresponding minimal motifs were identified by the western blotting analysis. And the conservation analysis indicated that ten of the nineteen minimal motifs were conserved among46PPPV strains. In addition, all of the randomly chosen linear B cell epitopes containing the19different minimal motif BCEs could react specifically with PPRV-Nigeria75/1goat serum. In the other study, the DNA vaccine expression vector pVAX-MSTN-Linker-PPRV-H of sheep was constructed in order to promote the muscle growth and resist PPRV.
