Carassius auratus in Qihe river was one ofthe local varied populations formed in the long-term ecological adaptation. The full-length cDNA cloning of myostatin gene frome Carassius auratus in Qiheriver.The full-length cDNA of myostatin gene of Carassius auratus in Qihe river was2094bp(Gene Bank:KC851952) which included a1128bp complete ORF encoding a375amino acidspeptide,a86bp long5’-untranslated region,and a880bp long3’-untranslated region.The375amino acids long putative peptide contains a22amino acids long signal peptide,a conserved RIRRproteolytic processing site,and9conserved cysteine residues in the Cterminal of the protein.Theclone of full-length myostatin of Carassius auratus in Qihe river was obtained RT-PCR and rapid-amplification of cDNA ends(RACE).The Carassius auratus in Qihe river MSTN had highsimilarity to the Cyprinus carpio myostatin in while had low similarity to the M.pellegrinimyostatin and lower similarity to the mammalian and bird myostatin.The phylogenetic analysesshowed that the Carassius auratus in Qihe river myostatin had closest relationship with theCommon carp myostatin.Phylogenetic analysis indicated that MSTN sequence homology amongvertebrate species correlated primarily to their relationship.2. Expression analysis of myostatin gene from Carassius auratus in Qihe river.And,myostatin mRNA tissue expression profile was assessed by real timequantitative-PCR.In the eight examined tissues,the myostatin gene was expressed strongly inbrain,weakly in muscle and kidney,least in intestine. The expression pattern of MSTN indicatedthat Carassius auratus in Qihe river MSTN could play important role in muscular development,and it could contribute to other biological functions.3. The histological comparison from back muscle tissue of four kinds of Cyprinidae fish.The morphological index date form four kinds of Cyprinidae fish with the same growthenvironment and the period show that:In terms of weight,the most was Silver carp and there wassignificant difference between the other three kinds of Cyprinidae fish.In the aspect of lengh ofbody, there was significant difference fromCarassius auratus in Qihe river and Cyprinuscarpio.Ctenopharyngodon idellus and silver carpt,but Ctenopharyngodon idellus and silver carptwere longer,In the high of body, Carassius auratus in Qihe river was the highest,which maintainthe consistency to the features"back high and body thickness"of Carassius auratus in Qiheriver.Compare the number of muscle fiber in unit area from back muscles fond that in contrastwith Carassius auratus in Qihe river,Ctenopharyngodon idellus was up to the number of four kinds of Cyprinidae fish. |
