Pinus densiflora,tall trees,can form the ectotrophic mycorrhiza. Ectotrophic mycorrhiza fungi are mostly edible fungus, tricholoma matsutake is one of them. But it is a pity that Tricholoma matsutake still cannot undertake artificial cultivation so far, tricholoma matsutake and Pinus are reciprocal symbiosis, the growth of tricholoma matsutake and Pinus must have a close relationship. This experiment is using DGGE technique, gene cloning and establish gene library method to analyse tricholoma matsutake, tricholoma matsutake rhizosphere and Pinus rhizosphere microbes. Provide a theoretical basis for the research of tricholoma matsutake and Pinus symbiotic relationship. The conclusions were shown as follows:1、We have established the PCR system of rhizosphere soil fungi of Pinus.25uL system contained template lOng, dNTPs0.15mM. the each primer3.2uM, Mg2+1.0mM,Taq enzyme2U.Besides we have selected for DGGE fingerprinting primers ITS1F(GC)-ITS2,341F(GC)-518R.2、Through tricholoma matsutake fruiting body and pinus rhizosphere microbes in PCR DGGE, obtained tricholoma matsutake fruiting body yield bacteria invasion, pinus ectotrophic mycorrhiza fungi and rhizosphere soil microbial diversity.3、Through the DGGE strip,cutting, get25bands, cloning, sequencing and BLAST.There are eight kinds of fungi in tricholoma matsutake fruiting body and pinus rhizosphere soil, respectively is Hypocrea sp, Tricholoma matsutake sp., Cenococcum geophilum sp., Russula sp., Inocybe sp., Dothideomycetes sp., Cortinarius sp, Ascomycete sp. all samples have Cenococcum geophilum sp.,there are16uncultured fungi.
