Cucumber (Cucumis sativus L.) is one of the most important vegetable crops cultivated world-wide. Gibberellin is plant hormones that is widespread. Bioactive gibberellin can promote plants various aspects growth that was synthesized throuth a series of enzymes. They are including GA20oxidases、GA2oxidases、GA3oxidases that are very important in the pathway of gibberellin biosynthetic. GA20oxidases are degrading enzymes that are responsible for degrading C20GA. GA2oxidases play the role of deactivateing the gibberellin. GA3oxidases can promote the synthesis of the bioactive gibberellin. Warty fruit trait is one of the highly valuable external quality traits of cucumber. The glabrousl mutant (gll) is showed no spine and tumor on cucumber fruit surface. Furthermore, gibberellin can promote the development of trichomes in Arabidopsis. In order to verify the role of GAoxidases in the process of formation of fruit spine in cucumber, detailed bioinformatics of cucumber GA20ox^GA2ox>GA3ox were analyzed. Genetic transformation was also conducted in cucumber and we have mediated genetic transformation experiment of GA20oxl and GA20ox5to define gibberellin affect the formation of fruit spines in cucumber. The main results were as follows:1. In this study,19cucumber GA20ox、GA2ox、GA3ox genes were identified in the latest cucumber genome. Phylogenetic analysis showed that the conserved motifs and gene structures of cucumber GA20ox、GA2ox、GA3ox genes show similarities with Arabidopsis.2. Tissue specific expression analysis showed that GA20ox、GA2ox、GA3ox have different patterns of roots, stems, leaves, male flowers, fruits and tendrils expression in cucumber.Furthermore, CsGA20ox1show high levels of transcript abundance in leaves and fruits.3. We have constructed GA20ox1、GA20ox5overexpression vectors and GA20oxl silencing vector in this study. Genetic transformation was also conducted in cucumber and we have got1335S:GA20ox1transgenic cucumber plants in T1generation. Furthermore we have found that the fruit spines of four35S:GA20ox1transgenic cucumber plants are shorter than WT in T1generation.In summary, we performed bioinformatics、RT-PCR、reverse genetics analyses of the GA20ox、GA2ox、GA3ox family in cucumber. We have got35S:GA20ox1genetically modified plants and we have found gibberellin affected the formation of fruit spines. The above results are lay the foundation for the further explorating the effection of gibberellin in the process of the formation of cucumber fruit spines.
