Panax ginseng belongs to Plantae, Angiospermae, Magnoliopsida, Apiales, Araliaceae,Panax. Ginseng is mainly distributed in the northeast of China and in Russia’s fareast, and isalso distributed in Japan, Middle Korea and North Korea. The medical bioactivities of ginsengwas studied as early as1845, and was found that the ginsenoside have the functions ofimmunoregulation, nutrition, sthenia, Anti-inflammation and etc.Ganoderma lucidum belongs to Fungi, Basidiomycota, Agaricomycetes, Polyporales,Ganodermataceae, Ganoderma. There are many species of Ganoderma in China, and mainlygrow in forest where the trees are sparse, and they are saprophytic fungi of broad leaf wood.Toth etl. first reported the antineoplastic activity of Ganoderma triterpenes (GTS) in1983, andsubsequent research found that Ganoderma lucidum have many medicinal values such as liverprotection, detoxification, antihypertension, cholesterol reduction, antioxidant, abirritation, etc.In order to find the optimum conditions of Ganoderma lucidum protoplast transformationwith electric shock. Using SQS gene of Panax ginseng as experimental materials, andtransformed it into Protoplast of Ganoderma lucidum through electric shock method. The bestvoltage was800V when the electrode spacing was2mm; The mot suitable electrode buffer was0.6M/L mannitol solution which contains0.1g/L CaCl2; The expected bands which consistentwith the left (699bp) and right (397bp) flanking sequences and SQS full longth section sequence(1245bp) were found, which preliminarily proved that the large fragment DNA had beentransformed into Ganoderma lucidum; The mycelia of positive transformation were still had theclamp-connection and less branches than the original ones. The mycelia of the transformatsgrowed faster and the margin was denser, the middle of mycelia was more crinkly. Theganoderic acid contents were increased.
