In this study, used16S rDNA PCR-RFLP, sequences analysis of16S rDNA,SDS-PAGE of whole cell protein and determination of soil nutrient indicators to stdudy thediversity of resources and phylogenetic on the130rhizobial isolates from China’s southerntropical and subtropical regions of Guangdong, Guangxi, Yunnan, Fuji an, Hainan.The results of16S rDNA PCR-RFLP show that all strains tested divided into threegroups at the similarity of64%. Group I belonging to the genera of Rhizobium, include onlytwo strains, including HBU53008and HBU53009. Group II belonging to the genera ofCupriavidus, the group divided into two subsets at the similarity of81%, subgroup1including HBU53006, HBU53027, HBU53028, et al., at the total of17strains, isolated fromNanNing, BeiHai, ZhanJiang, ZhuHai, DongGuan, WuZhiShan, Xi ShuangBanNa, Jinghongand XiaMen, subgroup2including HBU53007, HBU53016, HBU53017, et al., at the total of56strains, isolated from NanNing, HePu, BeiHai, ZhanJiang, GuangZhou, ZhaoQing,DongGuan, WuZhiShan, Jinghong, China-Burma border, XiaMen. Group III belonging to thegenera of Burkholdeira, the group divided into four subsets at the similarity of92%.Subgroup1get together with the reference stin of Burkholderia mimosatum LMG23256Tra,including24strains, isolated from Traditional Chinese Medicine, HePu, BeiHai, ZhanJiang,ZhuHai, DongGuan, NongDao, Xi ShuangBanNa, Jinghong and XiaMen, XingLong;subgroup2get together with the reference strain of Burkholderia phymatum LMG21445T,including22strains, isolated from Traditional Chinese Medicine, HePu, LeiZhou, ZhanJiang,Xi ShuangBanNa, FuZhou; subgroup3get together with the reference strain of Burkholderiacaribensis LMG18531T and Burkholderia sabiae LMG24235T, including a total of6strains,HBU51005isolated from ZhuHai, the remaining were captured from soil; subgroup4get togetherwith the reference strain of Burkholderia nodosa LMG23741T, including only3strains,isolated from LiuZhou.According to the result of16S rDNA PCR-RFLP, the16S rDNA sequencing analysis by16representative strains showed that the results of16S rDNA Sequencing analysis are ingood consistent with16S rDNA PCR-RFLP.16representative strains were distributed inthree phylogenetic branches according to the phylogenetic tree based on the16S rDNA genesequencesin, they are genera Burkholderia and Cupriavidus in b eta-Proteob acted a and thegenus Rhizobium in alpha-Proteobacteira. The result of SDS-PAGE of whole protein showed that all130tested strains divided into15different SDS-PAGE phenotypes. The two strains HBU53008and HBU53009in group Iof the16S rDNA PCR-RFLP have one SDS-PAGE phenotype.73strains in group II and55strains in group III all have seven different SDS-PAGE phenotypes. Have the sameSDS-PAGE phenotypes are not isolated from the same location, opposite the strains isolatesfrom the same place, they can produce different SDS-PAGE phenotypes.In rhizobium distribution research, the sampling point of Guangxin Province whereisolated the genus of Rhizobium groups, the nitrogen, phosphorus and potassium are in thelow state, the soil nutrition is very much lacking. The sampling points of Guangdong andGuangxi Province where isolated the genus of Burkholderia, the nitrogen nutirtion level at thesecond and third level are favor than the location isolateed the Cupriavidus groups; Thephosphorus nutritional status of sampling points of Guangdong and Guangxi Province are atthe level of the following four levels, the phosphorus nutirtion is lack or even lacking. Thesampling points of Guangxi Province where isolated the Burkholderia or Cupriavidusgroups, the potassium nutirtion are in the four and ifve levels, the potassium nutirtion is verylack. The sampling points of Guangdong Province where isolated the Burkholderia groups,the potassium nutrition are very rich, at the one or two levels, but the sampling points ofGuangdong Province where isolated the Cupriavidus groups, the potassium nutirtion at thelack of four and ifve level. The two sampling points of Guangdong Province not only isolatedthe Burkholderia genus but also isolated the Cupriavidus groups, the soil nutrition of nitrogen,phosphorus and potassium are at the lowest level of five and six. The soil nutrients are verylack, compared with the sampling point of Guangdong where isolated only one Rhizobiamgroups, only GD01sampling site is similar with it, the level of nitrogen, phosphorus andpotassium of the other sampling points are higher than the two sampling sites1-5levelrange.
