Preservatives as a kind of food additives are getting more and more attention with the increase of life level. Their types and dosages permitted were different in different countries. Benzoic acid and sodium benzoate which were banned in lots of countries were the most applied preservatives in China, but they were also forbidden in the fields of green food in A level. Parabens, sodium diacetate, sorbic acid and its potassium are safe and internationally recognized preservatives. Their toxic effects are lower than benzoic acid and sodium benzoate. Algal toxicity test of the three kinds of preservatives are seldom studied.Algal toxicity test of butylparaben sodium(BP), sodium diacetate(SDA) and potassium sorbate(PS) were studied in this paper. At algal physiological pH value (alkalescency condition), BP had no effect on Dunaliella tertiolecta with the concentration from 0 to 1.4×102mg/L. In the range of 1.5×102mg/L-2.3×102mg/L, the inhibition of BP to D. tertiolecta increased with the increase of concentration. Over the concentration of 1.7×102mg/L, BP had lethal effect on D. tertiolecta. Blow the concentration of 2.4×103mg/L, SDA had slightly promoting effect on D. tertiolecta; SDA has lethal effect on D. tertiolecta over the concentration of 2.4×103mg/L. Blow the concentration of 10g/L, PS had no effect on D. tertiolecta.Both PS and SDA take effect on acidic condition, so the pH of D. tertiolecta’s medium was adjusted to 4.2-4.5 and stays for 1 hour. The result showed that D. tertiolecta from logarithmic phase will reenter adjustment phase. In the 7th day, pH of D. tertiolecta’s medium increased to normal level and D. tertiolecta re-enter logarithmic phase, and there is no damage on D. tertiolecta during these period.when the medium pH of D. tertiolecta was adjusted to 4.2-4.5 and stayed for 1 hour, blow the concentration of 15.0mg/L,SDA had no effect on D. tertiolecta which was only affected by pH, but SDA has lethal effect on D. tertiolecta over the concentration of 16.0mg/L. Blow the concentration of 14.0mg/L, SDA had no effect on SOD activity of D. tertiolecta; SOD activity of D. tertiolecta decreases to a lower level if the SDA concentration was over 14.0mg/L. Blow the SDA concentration of 15.0mg/L, SOD activity of D. tertiolecta was only affected by pH, and CAT activity of D. tertiolecta was down to a lower level if the SDA concentration was overl5.0mg/L.Staying in acidic condition for 1 hour, D. tertiolecta was only affected by pH not PS blow the concentration of 3.0mg/L,and PS had inhibition to D. tertiolecta in the range of 4.0-7.0mg/L, and PS has lethal effect on D. tertiolecta over the concentration of 8.0mg/L. Both PS and pH had no effect on CAT activity of D. tertiolecta in the range of 0-4.4mg/L. SOD activity of D. tertiolecta decreases to a lower level with the concentration of PS in the range of 4.8-8.0mg/L.In the range of 0-4.8mg/L, PS had no effect on CAT activity of D. tertiolecta which was only affected by pH. In the range of 5.2-8.0mg/L, CAT activity of D. tertiolecta decreased with the PS concentration increasing, and CAT activity of D. tertiolecta is down to a lower level in the PS concentration of 8.0mg/L.The toxic effects of BP, SDA and PS to HL7702 cell also were studied in this paper. Blow the concentration of 62.5μ,g/mL, BP has no effect on HL7702 cell. In the range of 100-1000μg/mL,survival rates of HL7702 cell decreased from 87% to 11% with the increase of concentration, and EC50=215.97μg/mL. Blow the concentration of 1000μg/mL, SDA and PS had no effect on HL7702 cell, and the survival rates of cell were over 90%.
