Phellinus linteus is a kind of precious medicinal fungus, containing a variety of active ingredients, especially with the biological response modifiers (BRM) effect of active polysaccharides. Much attention has been paid to the research of active material of fungal polysaccharides, while at home and abroad, the study on the the application on hypoglycemic effect has little known, for the research and development of value of the hypoglycemic functional food and medicine. This paper studied the effect of Phellinus igniarius solid fermentation and liquid fermentation, mycelial polysaccharide from Phellinus igniarius polysaccharides, and traditional Chinese medicine compound in vivo hypoglycemic activity of polysaccharide, isolation and purification and structural basis for the exploration. The conclusions were as follows:1 For submerged fermentation the optimum carbon sources were corn powder and soybean powder, the best nitrogen sources were yeast extracted juice and soybean powder, the best carbon and nitrogen ratio was 25:1. The orthogonal experiments showed that the optimal conditions for submerged fermentation culture optimal conditions were:temperature 28℃, liquid volume 80 mL, rotating speed 110 r/min, fermentation time 9 d. It could be started with liquid fermentation of one or two strains, then transferring to the PDA cultured in a liquid culture medium to obtain uniform 5 d, transparent, moderate density of mycelial pellets.2 Choosed Ju Juncao as medium can make good economic benefit. The solid fermentation transfed to the liquid spawn, showed that the mycelial grew faster and could well reduce the period of culture.3 The results of single factor and orthogonal experimental optimization of mycelial polysaccharide extraction process showed that:with the condition of the extraction temperature 90℃, solid-liquid ratio 1:40, extraction time 1.5h, with 3 times volume ethanol alcohol polysaccharide, the effect was best with polysaccharide yield reaching 12.056%.4 Polysaccharide after deproteined, depigmentation, removing small molecules, with DEAE SephadexA-50 further purified and obtained three polysaccharide fractions, in turn named PIP-Ⅰ, PIP-Ⅱ, PIP-Ⅲ, PIP-Ⅰ SephadexG-200 purification, identification is homogeneous polysaccharide. The results of UV spectrum showed that PIP-Ⅰ without nucleic acid and protein fractions. Researched on the PIP-Ⅰ structure characteristics with Nuclear magnetic resonance hydrogen spectrum (NMR)and infrared spectral scanning.5 Combined with the compatibility of traditional Chinese medicine theory, using four alloxan-induced hyperglycemia in mice model of building, Phellinus igniarius mycelium polysaccharide and its compound Chinese medicine compound with hypoglycemic effect got the results that:the sample group of glycemic index and sugar tolerance index is positive; Phellinus igniarius compound polysaccharide and polysaccharide of Phellinus igniarius low dose group of lowering blood glucose had the best effect, compound of traditional Chinese medicine to some extent had auxiliary effect in reducing blood glucose.
