Jerusalem artichoke commonly known as Helianthus tuberosus linn, Yang Qiang is a perennial herb which is rich in SDF(soluble dietary fiber). Recently, the multifunctional abilities of SDF to reduce high blood pressure, reduce blood sugar and boost human immune system have been studied intensively. The main objective of this thesis is to extract SDF from Jerusalem artichokeusing composite enzymes and to evaluate the hyperlipidemic effects on hyperlipidemica mice.Furthermore, chewing tablets made from Jerusalem artichoke SDF was developed in order to provide a theoretical basis for product development and its application. This thesis contents and results are as follows:(1) Composite enzymatic extraction of SDF from Jerusalem artichokeFirst, the single-factor experiment, orthogonal experiment and response surface method were employed to compare the extracting ratio of differet enzymes and modified Jerusalem artichoke SDF, to optimize the extraction technology parameters. The Jerusalem artichoke SDF extracting parameters were tested to be optimum in the front stage:pectinase 0.25%, acid protease 0.2%, pH value 4.0, composite hydrolysis temperature 50℃, reaction time 3h, solid-liquid ratio 1:10g/ml,and in the last stage:cellulase0.52%, hydrolysis pH value of 5.91, reaction temperature 57.71℃, hydrolysis time 1.82h. and under this optimal technology condition the extraction ratio went up to 9.53 percent. Before the modification and the modified difference between water-holding capacity, oil holding capacity and viscosity of the SDF’s very significant, and increased by 1.01 times,1.19 times and 2.70 times.(2)the hyperlipidemic effect of Jerusalem artichoke SDF on the hyperlipidemia miceAfter the hyperlipidemia mice was treated with Jerusalem artichoke water-SDF, the indicators of the serum and liver tissue of mice were generally improved, liver tissue damage was mitigated, the vitality AST and ALT in serum were significantly decreased. The effects Jerusalem artichoke SDF on serum and liver tissue indicators were distinguished by defferent SDF dosage through the comparison of TC, TG, HDL-C and AI model.The resoults showed that the lower and higher dosage were proved to be inferior to medium dosage of SDF. Therefore, Jerusalem artichoke SDF can be use to remedy hyperlipidemia and promote the function of lipid metabolism.(3) The development of Jerusalem artichoke SDF chewing tabletsThe optimal formula and technology parameters for Jerusalem artichoke SDF chewable tablets was explored and determine as follows:Jerusalem artichoke SDF 22%, mannitol 18%, apple powder 22%, maltodextrin 8%, comstarch 10%, β-cyclodextrin 15-20%, magnesium stearate 1%, dried at 65℃ for 40min, the granulation humectant with 75% ethanol. The tablet hardness were influenced by the addition of maltodextrin, corn starch and microcrystalline cellulose. The addition of maltodextrin 8%, corn starch 5% or microcrystalline cellulose 20% were tested to be compatible for the tablet hardness, while β-cyclodextrin gets insignificantly effect on the tablet hardness.
