Sheep bone powder was hydrolyzed by alcalase and the antioxidative activity of the resulted Ossein Oligopeptides (OOP) was investigated for the future high-value utilization of sheep bone.Obtaining rate of short peptides was used as the indicator during the optimization of hydrolyzing conditions via responsive surface methodology.When the substrate was hydrolyzed under the resulted optimized condition,that is,8.24% substrate,4.35% alcalase,46℃ for 5h at pH 9,the obtaining rate of OOP reached to 71.19%,which is quite close to the predicted value of the model. In vivo study suggested that 0.6 mg/mL OOP exhibited higher ·OH scavenging ability than 0.3mg/mL ascorbic acid(Vc) and O.lmg/mL lipoic acid(P<0.05). 0.2mg/mL OOP exhibited higher DPPH-scavenging rate than 0.2mg/mL Vc and O.lmg/mL lipoic acid. At the concentration of 0.4 mg/mL, OOP showed higher O2-·scavenging ability than 0.1mg/mL Vc (P<0.05). In vivo study demonstrated that low-dose OOP increased the SOD and GSH-Px activity, and lowered MDA amount in the serum and liver significantly in a dose-dependant fashion. In conclusion, collagen short peptides prepared by alcalase has antioxidant activity both in vivo and in vitro and can be widely used in food, medicine and other areas.
