In this paper, carp skin was used as raw material to study the conditions of extracting collagen from carp skin, the preparation process of cross-linked collagen and some properties of cross-linked product. The main process and results are as follows:1.The process conditions of carp skin collagen were determined. With acetic acid 2.0%, the solid-liquid ratio of skin(W) and acetic acid(V) 1:40 and soaking time 180 min, the extraction rate of skin collagen was 60.0%. Amino acid composition, SDS-PAGE pattern and fourier transform infrared spectroscopy showed that the collagen was I type collagen with the maximum absorption wavelength of 234 nm.2.The preparation process of collagen-chitosan cross-linked products using glutaraldehyde as a crosslinking agent was studied. The crosslinking reaction conditions determined by orthogonal experiment were: the composition ratio of collagen and chitosan 6:4, the total concentration of collagen and chitosan 0.8%, the concentration of glutaraldehyde 2.0%, the absorbent multiples of cross-linked product was 48.4 and the tensile strength 100.5 N/mm2. The coagulation index was 23.0 and cross-linked product was spongy solid after lyophilization.3.The physicochemical properties of cross-linked product were analysed. The degree of crosslinking of cross-linked product was 33.8% with apparent density 0.0125 g / mm3 and porosity 78.0%.4.By contrast with the absorbent multiples, tensile strength and coagulation index of gauze,collagen-chitosan cross-linked products without glutaraldehyde, commercially available gelatin hemostatic sponge, the present cross-linked product showed excellent properties which reflected that the carp skin collagen could be used as the raw material of hemostasis cross-linked product.
