Nannochloropsis salina is a single-celled algae. because it is rich in protein, polyunsaturated fatty acids and pigments, it is considered to be the high added value of biological. And the simple culture conditions, no consumption of freshwater resources, and easy to large-scale farming, no doubt has great business development potential. Transcription factors(TFs: transcription factors) is a kind of protein molecules. It could identify and bind the cis-acting elements of promoter region specifically to activate or inhibit the transcriot of genes.In this experimental study,the gene sequence of NgAUREO1 was from the laboratory preservation. It was constructed the yeast expression vector pAUR101-AUREO, Which was successful transformed into Saccharomyces cerevisiae INVScI by electroporation successfully and binded to Saccharomyces cerevisiae chromosome by homologous recombination. After detection by PCR amplification and RT-PCR, AUREO was successfully expressed in DNA, RNA level. According to the characteristic of AUREO identification and bind to DNA core sequence(TGACGT), we selected 4 genes related with cell growth(ATP1、GUA1、PBS2、BNA3). Analysis of expression of each gene by RT-PCR technology, we found that there are 2 genes have significantly changed, which ATP1 play the expression increased, PBS2 decreased expression.This study make us further understand the AUREOs functionality and lay the foundation for applying AUREOs in genetic modification to control gene expression in vivo.
