| The number of patients with alopecia has been growing rapidly, and the Age has been trending younger. Now most of the researches on transplant hair are based on the patients with enough hair at home and abroad, but there was no apparent corrective effect for large area of alopecia. Implanted artifical hairs could solve this problem.PET and polypyrrole were used to make PPy/PET composite material by liquid in-situ polymerization method. Since PET has good biostability and great biocompatibility. PPy has stable characteristics, good flexibility, good biocompatibility and strength, PPy can heighten PET monofilament biocompatibility to meet the need of artificial hair implantation, which can be a new therapeutic method for alopecia.To make and evaluate the KH-550/PPy/PET composite monofilament as implanted artificial hairs, and carry out cytotoxicity tests using NCTC clone929cells with composite monofilament.KH-550/PPy/PET composite monofilament have been prepared in a series of steps, which include of pre-spotting, alkali treatment, silane coupling agent treatment and polypyrrole coating.The viability of NCTC clone929cells are detected after1,2,3,5,7days culture.PET material, collagen materials, PPy/PET composite material and KH-550/PPy/PET composites were transplanted into the alopecia area of New Zealand white rabbits. Biological stability were analysised by regularly observing the apparent shape and organization of composite monofilament material, HE staining and scanning electron microscopy.KH-550/PPy/PET composite monofilament has a smooth surface without crack. The PPy films won’t come off accidentally and have good wearability. After alkali treatment, PPy’s quality on the surface of monofilament is significantly heavier than before. Using silane coupling agent (KH-550) could effectively enhance biocompatibility and binding force of polyester monofilaments. NCTC clone929cells were cultured with monofilaments together.The livability of cells is82.63%ã€80.37%ã€73.26%ã€81.96%ã€77.50%by time respectively. The results showed that the level of cytotoxicity was grade1and there is no obvious cytotoxicity and no acute toxicity of materials.HE staining biopsy results showed that the surrounding tissue of all groups with implanted material appeared inflammatory cells at early period, and almost were at the same degree of inflammation. After one month, the inflammatory cells around the Collagen group, PPy/PET composite group and KH-550/PPy/PET composite group can be seen less than PET material groups. After two months, there were almost no inflammatory cells around each group and can observed connective tissue wrapping material surface. Composites have good biocompatibility. |
