Menispermi Rhizoma (Chinese name Bei Dougen) is the dried rhizoma of Menispermum dauricum DC.. In the record of Chinese Pharmacopoeia (2010), it has used in treating swollen sore throat, dysentery, rheumatism and toxic-heat. The major components in Menispermi Rhizoma, alkaloids, display diverse biological activities, including antiarrhythmic, antibacterial, anti-inflammatory, anti-hypertension, antineoplastic and inhibition of platelet aggregation, which are indicated by modern pharmacological research. We researched on the chemical components and pharmacological activity of Menispermi Rhizoma, which may be valuable for Menispermi Rhizoma further studies and utilization.Menispermi Rhizoma was firstly extracted with75%ethanol by reflux, after evaporated the solvent, the dryness of the extracts were subjected to conventional column chromatography to separate the components. Seven compounds were separated and identified by TLC, MS, UV, IR, H’NMR and C13NMR methods. They are tetrandrine, dauricine, daurisoline, daurinoline, sinomenine and acutumine. In addition, the HPLC method was used for quantitative analysis of five main alkaloids in Menispermi Rhizoma. This could be used as quality control method of Menispermi Rhizoma.We chose the ulcerative colitis model induced by TNBS in mice and used this model to evaluate the anti-inflammatory effects of total alkaloids of Menispermi Rhizoma. The results indicated that in comparation with model group, different dose of Menispermi Rhizoma alkaloids (MDA) could significantly decrease MPO, TNF-a, IL-1β and IL-6colon tissue levels and the expression of COX-2. And MDA could also down-regulate the NF-κB/TLR4mRNA expression in colon. Moreover, MDA repaired the macroscopic damage and the histopathological changes in colon after TNBS induction. All the parameters results showed the protective effect of MDA in TNBS-induced ulcerative colitis. In summary, MDA may have therapeutic value towards inflammatory bowel disease. |
