| The2010edition of "Chinese Pharmacopoeia" stipulate Astragalus:the dried root of Astragalus Mongholicus Bunge (Astragalus membranaceus (Fisch.) Bge. var. mongholicus (Bge.) Hsiao) and Astragalus membranaceus (Astragalus membranaceus (Fisch.)) was used as genuine medicinal. Astragalus is widely used in our proprietary proprietary Chinese medicine preparations to enhance immune function, hepatoprotective, diuretic, anti-aging, anti-stress, buck and broader antibacterial effect. There is a wide variety of preparations containing Astragalus, including Buzhongyiqi pills, Ejiaobuxue oral solution, Zhenqifuzheng particles, Yiganning particles, Yupingfeng dripping pills and and so on. The existing quality standards of Radix Astragali and the preparations containing Astragalus are more than to determine the content of Astragaloside by HPLC-ELSD.The subject specific research mainly includes the following aspects:1. To inspect the transfer rate of three Astragalus preparations including Buzhongyiqi pills, Ejiaobuxue oral solution, Zhenqifuzheng particles by HPLC-ELSD method. The Astragaloside transfer rate of Buzhongyiqi pills is significantly higher, up to400%; Ejiaobuxue oral solution is95%; Zhenqifuzheng particles is30%, which is relatively low. Buzhongyiqi pills is raw powder medicine, the preparation method is relatively simple.To study the Astragaloside metastasis on all aspects of semi-finished and finished products and examine Astragaloside transfer rate reasonableness. 2. Through four astrageloside structure analysis, using liquid chromatography separation, Q-Tof mass spectrometry to study the stability and mutual transformation of Astragalus in saponins, and research the interference factors of influencing astragaloside Determination.The results confirmed that Astragalus saponin I, II can be converted into Astragaloside in an alkaline environment, Astragalus saponin III does not occur conversion, acidic environment can produce the damaging effects of Astragalus Saponins I, II, III, IV, making their contents decreased. This study provides effective research base for the quality control of Astragalus herbs as well as the preparations containing Astragalus.3. UPLC-MS method was used to determine the content of four active ingredients including Astragalus saponin I, II, â…¢, IV (Astragaloside). Through multi-components quantitation to evaluate the quality of Radix Astragali, and to exploratory study the method of multi-components quantitive by one marker. Through the above research to establish a more accurate and reliable quality control program and methods of Radix Astragali and its preparations, to make their drug quality standards rationalization and scientific. |
