Jiaweisimiao’s Fingerprint Of Effective Elements And Therapeutic Basis

The purpose of this study is to understand the therapeutical basis of Jiaweisimiao Decoction.By the research, we can strengthen the understanding of compatibility, and find the therapeuticalbasis of Jiaweisimiao Decoction.The research does a systematic studies to effective compositions and establishes the basis forexploitation of a new drug.The research concludes experimental design using orthogonal table and valid targets’extraction、separation、purification; the drug effect research; the building of valid targetsfingerprint;TCM chromatographic pharmacodynamics; the attribution research of fingerprintpeaks.First of all, we get 6 valid targets by extraction, separation and purification from JiaweisimiaoDecoction. They are alkaloids、flavones、oils、organic acids、saponins and sugers. We combinethem according to orthogonal design. Then, we get a few examination.And then, we carry through pharmacodynamics to these. We choose analgesia、antiinflammatory action、descending uric acid as the drug effect indexes according to the analysis togouty. We found the functionary valid targets by the drug effect indexes.We used effective fraction cluster (concluding 6 valid targets)to build HPLC and GCfingerprint. The effective fraction cluster conclude a great variety of different chemical propertycompositions. So we must use HPLC and GC to analyze effective fraction cluster for moreinformation.Through the retention time of fingerprint peaks and the ultraviolet spectra of peaks acquiredby DAD, we can confirm HPLC fingerprint peaks’ attribution. And we analyze oils’ compoundsby GC-MS. Then, we compare sugers acetylation with drug acetylation to confirm C4 fingerprintpeaks’attribution.We adopted multiple regression analysis(enter and stepwise)and correlation to look for therelation at fingerprint peaks and drug effect. We found many fingerprint peaks they haveinfluences to drug effect by these statistical methods.Conclusion: we built valid targets fingerprint, found many fingerprint peaks they haveinfluences to drug effect and confirm fingerprint peaks’ attribution by above a series ofresearches. This is a attempt to find TCM therapeutical basis.
