Objective:Pyridinoline crossed-linked carboxyterminal telopeptide of type I collagen (ICTP) is a specific marker for bone resorption. Early studies have showed strong correlation between ICTP levels in gingival crevicular fluid (GCF) and severity of alveolar destruction among patients with periodontitis. Calprotectin (S100A8/A9complex) is regarded as a symbol for inflammatory diseases. The high expression of GCF calprotectin is closely related to the extent of periodontal inflammation. The aim of this study was to detect GCF levels of ICTP and S100A8/A9before and after periodontal therapy, so as to explore whether they could be used together as biomarkers in the diagonosis and evaluation of treatment effect of periodontitis.Methods:14chronic periodontitis and8healthy people were enrolled. A total of28GCF samples from experimental group and16samples from control group were collected in the mesial or distal site of molars with absorbent paper points. The weight of GCF and clinical parameters including gingival index (GI),probing depth (PD) and attachment loss (AL) were recorded.14patients received periodontal initial therapy and GCF samples were collected again after one week. ICTP and S100A8/A9levels in GCF were quantified using Enzyme-linked immunosorbent assay (ELISA). Data were analyzed statistically.Results: 1. ICTP and S100A8/A9concentrations in GCF from chronic periodontitis before therapy were higher than control groups respectively. The differences were statistically significant (P<0.01).2. GCF concentrations of ICTP and S100A8/A9were positively correlated with GI,PD and AL before therapy (P<0.05).3. ICTP and S100A8/A9concentrations decreased significantly one week after initial therapy,and were still higher than control levels (P<0.01).4. GI, PD and AL decreased significantly after initial therapy (P<0.01).5. There were positive correlations between the ICTP concentration and the S100A8/A9concentration both before and after initial therapy (P<0.01).Conclusion:The ICTP and S100A8/A9levels in GCF from chronic periodontitis are higher than in healthy people and have positive correlations with clinical indexes. After periodontal initial therapy,they decrease significantly along with improvement of clinical indexes. Moreover, ICTP and S100A8/A9levels have obvious positive correlations. The results indicate that ICTP and S100A8/A9can be used together as supplementary biomarkers in the diagnosis and monitor of treatment response of periodontitis.
