Object The research combines hyperhomocysteinemia triggered by high metfionine diret with atherosclerosis caused by balloon injury of the abdominal aorta. This study aimed to investigate the effects and mechanism of Hippophae Flavone on Atherosclerosis in rabbits induced by hyperhomocysteinemiaive.Methods Apart from the control group, rabbits in other groups underwent balloon catheter injuries and a high methionine diet to induce hyperhomocysteinemia and atherosclerosis. Forty eight male rabbits were randomLy divided into six groups:a control group(constant volume of normal saline), model group(constant volume of normal saline), Low-(0.32g/kg)、middle-(0.76g/kg) and high-dosage (1.6g/kg) Hippophae Flavone groups, western medicine [folic acid (0.58mg/kg), VitBg (1.15mg/kg) and VitB12(28.75ug/kg)] group.Rabbits were given relevant medicines intragastrically once a day for consecutive eight weeks. Twenty weeks later, the Hey concentration was measured by high performance liquid chromatography. With HE staining, we observed artery morphology alteration. The expression of MCP-1and NF-κB in aorta was examined by immunohistochemistry.Results After twenty weeks the plasma homocysteine concentration of the model group were significantly higher the control group, Both Hippophae Flavone groups and western medicine group could decrease the level of plasma Hcy (P<0.01). Aortic intimal changes in pathomorphology:in the western medicine group and Hippophae Flavone groups were attenuated compared with the model group, while the levels of the NF-κB expression in aorta of Hippophae Flavone groups and western medicine group were lower compared with the model group (P<0.01).TFH groups markedly reduced the expression of MCP-1in aorta endothelial cell detected by immunohistochemistry (P<0.01), The efficacy of Hippophae Flavone at a high dose was superior to the western medicine group (P<0.01).Conclusion (1) High methionine diet combined with balloon injury may cause the atherosclerosis.(2) Hippophae Flavone reverses aortic lesions induced by Hey.(3) The mechanisms may be related to its suppressive effects on NF-κB activation and MCP-1expression.
